摘 要:为了探究黑藏羊心脏在不同发育时期的氧适应相关通路和基因,本试验对青海省海南藏族自治州贵南县黑藏羊的心脏组织进行转录组测序。按照呼吸方式的差异设置了三个组:胚胎期(胎内气体交换)、羔羊期(肺呼吸)和成年期(肺呼吸)。利用京都基因与基因组学百科全书(KEGG)和基因本体(GO)数据库对差异表达基因的富集途径进行分析。通过STRING以及分子复合物检测算法(MCODE)进行蛋白质—蛋白质相互作用(PPI)网络的功能注释和构建。采用实时荧光定量PCR对关键基因SKP2、BUB1、CDC20和CDK2的表达情况进行检测。结果表明:富集在细胞周期、细胞粘附和细胞免疫等通路的基因在羔羊期和成年期显著下调。从差异表达基因的PPI网络和MCODE中鉴定出129个节点,在富集最显著的2个模块分别鉴定出41和20个核心节点基因。说明黑藏羊心脏的低氧适应与细胞周期、细胞粘附和细胞免疫等通路密切相关。SKP2、BUB1、CDC20和CDK2等基因在氧适应过程中发挥重要作用。In order to explore the pathways and genes related to oxygen adaptation in the heart of black Tibetan sheep at different developmental stages,transcriptome sequencing was performed on the heart tissue of black Tibetan sheep in Guinan,Hainan Tibetan Autonomous Prefecture,Qinghai Province.In this study,three groups were set up according to the differences in breathing patterns:embryonic period(intra-fetal gas exchange),lamb period(lung respiration)and adult period(lung respiration).The enrichment pathways of differentially expressed genes were analyzed using the Kyoto Encyclopedia of Genes and Genomics and Gene Ontology databases.The protein-protein interaction network was annotated and constructed by STRING and molecular complex detection algorithm.The expression of key genes SKP2,BUB1,CDC20 and CDK2 was detected by real-time fluorescence quantitative PCR.The results showed that the genes enriched in cell cycle,cell adhesion,cell immunity and other pathways were significantly down-regulated after the birth of black Tibetan sheep.Identified 129 nodes from the PPI network of differentially expressed genes and MCODE,and 41 and 20 core node genes were identified in the two modules with the most significant enrichment.This experiment shows that the hypoxia adaptation of black Tibetan sheep heart is closely related to cell cycle,cell adhesion,cell immunity and other pathways.SKP2,BUB1,CDC20,CDK2 and other genes play important roles in the process of oxygen adaptation.
