摘  要:【目的】探究短期关停补水工程对筼筜湖水质及水体中微生物群落结构的影响,为筼筜湖进一步综合治理提供新思路。【方法】以关闸和开闸期间的筼筜湖水为样本,测定其水质指标;采用高通量测序技术,分析两个时期水体样本中微生物多样性和群落结构的差异;同时,利用冗余分析(RDA)方法分析关键差异微生物与水质指标的相关性。【结果】与开闸期间比较,关闸期间水质指标的变化表现为:pH值、活性磷酸盐含量、氨氮含量、非离子氨含量明显升高,化学耗氧量(COD_(Mn))略微升高,无机氮含量下降。关闸期间水体中微生物多样性下降。经层层筛选,获得13个差异显著的操作分类单元(OTU),其中,关闸期间有12个对应的关键微生物在属水平的相对丰度高于开闸期间,且这些微生物与pH值、氨氮含量、COD_(Mn)和活性磷酸盐含量呈正相关。【结论】短期关闸停止了水体交换,水体运行动力不足,引起水体中活性磷积累明显、有机质转化不良、氨氮和非离子氨含量急剧上升等现象,提升了局部水质恶化的风险。【Objective】The effect of short⁃term closure of Yundang Lake replenishment project on water quality and microbial com⁃munity structure was investigated to provide new ideas for further comprehensive management of Yundang Lake.【Method】Water was sampled during sluice closure and opening for the determination of water quality indexes;differences in microbial diversity and com⁃munity structure in water samples from the two periods were analyzed by high⁃throughput sequencing technology;meanwhile,redun⁃dancy analysis(RDA)was used to analyze the correlation between the key differential microorganisms and water quality indexes.【Result】During the sluice closure period,the waters had significantly higher pH value,greater contents of active phosphate,ammo⁃nia nitrogen and non⁃ionic ammonia,slightly elevated chemical oxygen demand(CODMn),lower inorganic nitrogen content and reduced microbial diversity compared with the counterparts during sluice opening. Furthermore, 13 operational taxonomic units(OTUs) with significant differences were screened;among them the relative abundances of 12 microorganisms at the genus levelwere higher during sluice closure period than the open period, and these microorganisms were positively correlated with pH value,CODMn and contents of ammonia nitrogen and active phosphate. 【Conclusion】The short⁃term closure of sluice stops water exchange,and gives rise to insufficient running water power, accumulation of active phosphorus, poor conversion of organic matter, and dra⁃matic rise of ammonia nitrogen and non⁃ionic ammonia content, which increases the risk of regional water deterioration.
