摘 要:目的:研究牛大力总黄酮的大孔树脂纯化工艺及其抗氧化活性。方法:筛选对牛大力总黄酮吸附量最大的树脂,优化总黄酮的动态吸附、水洗除杂和动态解析工艺,采用ABTS^(+)·、DPPH·和·OH法测定牛大力总黄酮的抗氧化活性。结果:采用AB-8树脂,上样液总黄酮浓度为0.34 mg/ml,流速为1.0 ml/min,静置12 h后用2 BV水洗除杂,最后用3 BV 75%乙醇解析,乙醇流速为5.0 ml/min,纯化后总黄酮平均含量为75.86 mg/g。与纯化前比较,纯化后的牛大力总黄酮对·OH的清除能力明显增强(P<0.01),而对ABTS^(+)·、DPPH·的清除能力影响不明显(P>0.05)。结论:采用AB-8树脂纯化牛大力总黄酮工艺简单、稳定可行,纯化后的总黄酮抗氧化活性得到提高。Objective:To study the purification process of total flavone of Nanhaia speciosa(TFNS)using macro⁃porous resins and evaluate the antioxidant activity of the purified total flavone.Methods:The resin with the highest ad⁃sorption capacity for TFNS was selected.The dynamic adsorption,impurity removal by water washing,and dynamic elu⁃tion processes were optimized.The antioxidant activity of the purified total flavone was determined using the ABTS^(+)·,DPPH·,and·OH radical scavenging methods.Results:AB-8 resin was used in the purification process.The total fla⁃vonoid concentration of the sample loading solution of TFNS was concentrated to 0.34 mg/ml and passed through the res⁃in column at a flow rate of 1.0 ml/min.After static adsorption for 12 h,impurities were removed with 2 bed volumes(BV)of water.The purified total flavone were eluted with 3 BV of 75%ethanol at a flow rate of 5.0 ml/min.The content of to⁃tal flavone after purification was 75.86 mg/g.Compared with the pre-purified sample,the scavenging ability of the puri⁃fied total flavone against·OH radicals was significantly enhanced(P<0.01),while the scavenging abilities against ABTS^(+)·and DPPH·radicals were not significantly affected(P>0.05).Conclusion:The purification process of TFNS us⁃ing AB-8 resin is simple,stable,and feasible.The antioxidant activity of the purified total flavone is improved.
