摘  要:目的探讨糖尿病认知障碍大鼠脑组织形态学变化及生长相关蛋白(GAP)-43蛋白表达变化。方法雄性SD大鼠随机分为实验组和对照组,实验组腹腔注射链脲佐菌素(STZ)构建1型糖尿病模型,Morris水迷宫实验评价大鼠学习记忆能力,苏木素-伊红(HE)染色观察脑组织形态学变化,Western印迹和免疫组织化学(IHC)法检测GAP-43蛋白表达变化。结果实验组较对照组空腹体质量增长明显缓慢,空腹血糖明显升高(P<0.01);Morris水迷宫实验中,相较于对照组,实验组平均逃避潜伏期存在显著差异,穿越平台次数明显减少(P<0.05,P<0.01);HE染色显示,对照组脑组织神经元结构完整、形态正常,实验组脑组织锥体细胞数量减少、形态发生变化;Western印迹和IHC结果显示,实验组较对照组脑组织中GAP-43蛋白表达水平明显降低(P<0.01)。结论糖尿病大鼠出现认知功能障碍可能与GAP-43蛋白表达降低有关。Objective To investigate the changes of brain histomorphology and protein expression of growth-related protein(GAP)-43 in diabetic rats with cognitive impairment.Methods Male SD rats were randomly divided into experimental group and control group.The experimental group was injected with streptozotocin(STZ)intraperitoneally to construct type 1 diabetes model.Morris water maze experiment was used to evaluate the learning and memory ability of rats,hematoxylin-eosin(HE)staining was used to observe the morphological changes of brain tissue,and Western blot and immunohistochemistry(IHC)method were used to detect the changes in the expression of GAP-43 protein.Results Compared with the control group,the growth of fasting body mass was significantly slowed,fasting blood glucose was significantly increased in the experimental group(P<0.O1).In the Morris water maze experiment,compared with the control group,the average escape latency was significantly different,and the number of crossing the platform was significantly reduced in the experimental group(P<0.05,P<0.01).HE staining showed that the structure of neurons in the control group was complete and the shape was normal,while the number of pyramidal cells in the experimental group was reduced and the shape was changed.Western blot and IHC results showed that the expression level of GAP-43 protein in the brain tissue of the experimental group was significantly lower than that of the control group(P<0.01).ConclusionssCognitive dysfunction in diabetic rats may be related to the decrease in GAP-43 protein expression.
