摘 要:【目的】研究安徽省黄连木种质资源的遗传多样性,旨在为安徽省黄连木种质资源的保护利用和品种选育提供理论依据和实践指导。【方法】运用4对AFLP多态性有效引物对16个居群共341个样本单株进行遗传多样性和遗传结构分析评价,计算各遗传多样性参数,进行分子方差分析(AMOVA),对居群进行非加权算数平均配对法(UPGMA)聚类分析和主坐标分析(PCA)。【结果】1)共扩增出652个多态性位点。在物种水平上,多态性位点百分率(PPL_(s))为100%,有效等位基因数(N_(es))为1.233,Nei’s基因多样性指数(H_(es))为0.182,Shannon信息指数(I_(s))为0.323;在居群水平上,平均多态性位点百分率(PPL_(p))为52.15%,有效等位基因数(N_(ep))为1.214,Nei’s基因多样性指数(H_(ep))为0.137,Shannon信息指数(I_(p))为0.218。2)二层次和三层次的AMOVA分析均显示,黄连木遗传变异主要存在于居群内。区域(皖北、皖中、皖南、大别山区)之间遗传差异不显著;居群间遗传分化系数(G_(st))为0.243,基因流(N_(m))为1.562,UPGMA聚类分析将16个居群分为4组,与主坐标分析结果一致。研究还发现了一些种质特异性AFLP标记。【结论】安徽省黄连木种质资源的遗传多样性与居群间遗传分化均呈现较高水平,有必要采取综合措施来保护黄连木的遗传多样性。【Objective】The genetic diversity of germplasm resources of P.chinensis in Anhui Province was investigated,in order to provide theoretical basis and practical guidance for conservation,utilization and tree breeding of germplasm resources of P.chinensis in Anhui Province.【Method】The genetic diversity and genetic structure were evaluated for 341 sample trees from 16 populations of the species based on four polymorphically effective AFLP primer pairs.The calculation of genetic diversity parameters,analyses of molecular variance(AMOVA),population clustering analyses based on unweighted pair group method using arithmetic averages(UPGMA)and principal coordinates analyses(PCA)were conducted.【Result】1)A total of 652 polymorphic loci were detected.At the species level,the percentage of polymorphic loci(PPL_(s))was 100%,the effective number of alleles(Nes)was 1.233,Nei’s gene diversity(H_(es))was 0.182,and the Shannon’s information index(I_(s))was 0.323;At the population level,PPLp was 52.15%,Nep was 1.214,Hep was 0.137,I_(P) was 0.218.2)AMOVA at two and three hierarchical levels revealed that most genetic variation resided within populations.There is no significant genetic differentiation among geographic regions(Northern Anhui,Central Anhui,Southern Anhui and Dabieshan Moutains);The genetic differentiation among populations(G_(st))was 0.243,gene flow among population(N_(m))was 1.562.The UPGMA dendrogram displayed that 16 populations were divided into four major groups,which was consistent with the results of PCA analysis.Some germplasm-specific AFLP markers were found also in the present research.【Conclusion】The genetic diversity within populations and differentiation among populations of germplasm resources of P.chinensis in Anhui Province showed a relative high level.Conservation of germplasm resources of P.chinensis in Anhui Province with comprehensive measures is necessary.
