摘 要:目的探讨拔罐治疗银屑病大鼠的生物力学机制。方法取13只SD大鼠,将其随机分为正常组、模型组和拔罐组。采用5%咪喹莫特乳膏(IMQ)建立银屑病大鼠模型;采用万能材料测试机测试皮肤拉伸力;采用酶联免疫吸附测定(ELISA)法检测大鼠血清中白细胞介素-6(IL-6)、IL-17、IL-23及肿瘤坏死因子-α(TNF-α)的含量;采用实时荧光定量核酸扩增检测法(qPCR)检测大鼠皮肤组织中IL-6、IL-17、IL-23及TNF-α的含量以及核因子-κB(NF-κB)p65的mRNA表达水平;采用蛋白质印迹法(WB)检测皮肤组织NF-κBp65和磷酸化Smad2(pSmad2)蛋白的表达量。结果3组大鼠的皮肤拉伸强度差异有统计学意义,拔罐组明显高于模型组(P<0.05);与模型组比较,拔罐组大鼠的银屑病面积和严重程度指数(PASI)评分改善优于模型组(P<0.05),拔罐组大鼠的皮损厚度低于模型组(P<0.05);ELISA检测结果显示3组大鼠血清中IL-6、IL-17、IL-23和TNF-α的含量比较差异无统计学意义(P>0.05);qPCR检测结果显示,3组大鼠皮肤组织中IL-6、IL-17、IL-23和TNF-α的含量差异有统计学意义(P<0.05),拔罐组大鼠明显低于模型组(P<0.05);拔罐组大鼠皮肤组织中NF-κB p65的mRNA表达量明显低于模型组(P<0.05);拔罐组大鼠皮肤组织中NF-κB p65和pSmad2蛋白水平低于模型组(P<0.05)。结论拔罐可以增加银屑病大鼠皮肤弹性,减轻皮损局部炎性反应,拔罐可能通过抑制NF-κB信号通路改变了细胞的生物学效应。Objective In order to explore the biomechanical mechanisms of cupping in the treatment of psoriasis in rats.Methods Fifteen Sprague-Dawley(SD)rats were taken and randomly divided into the normal group,the model group and the cupping group,with five rats in each group.The psoriasis rat model was established by applying 5%imiquimod ointment(IMQ)cream.The tensile strength of the skin was tested by a universal material testing machine.The levels of interleukin-6(IL-6),IL-17,L-23 and tumor necrosis factor-α(TNF-α)in rat serum were detected by enzyme-linked immunosorbent assay(ELISA),and the levels of IL-6,IL-17,IL-23,TNF-αand the messenger RNA(mRNA)expression level of the transcription factor nuclear factor-κB(NF-κB)p65 in rat skin tissue were detected by quantitative polymerase chain reaction(qPCR).The relative expression of local transcription factors NF-κB p65 and pSmad2 proteins in skin lesions was detected by Western blotting(WB).Results The differences in skin tensile strength among the three groups of rats were significant(P<0.05),and the cupping group was significantly higher than the model group(P<0.05);compared with the model group,the improvement in the PASI scores of the rats in the cupping group was better than that of the model group(P<0.05),and the thickness of skin lesions of the rats in the cupping group was lower than that of the model group(P<0.05).The results of ELISA showed no significant differences in the serum levels of IL-6,IL-17,IL-23 and TNF-αin the three groups of rats(P>0.05).According to the qPCR results,there were statistically significant differences in the levels of IL-6,IL-17,IL-23,and TNF-αin the skin lesion tissues of the three groups of rats(P<0.05).Moreover,the rats in the cupping group had significantly lower levels than the model group had.The mRNA expression level of NF-κB p65 of rats in the cupping group was significantly lower than that in the model group(P<0.05),and the levels of NF-κB p65 and pSmad2 proteins in the skin lesion tissues of rats in the cupping group
