复方凉血消银饮对银屑病合并肥胖模型小鼠IGF-1水平影响的研究

摘　　要：目的探讨复方凉血消银饮对银屑病合并肥胖模型小鼠胰岛素样生长因子-1(IGF-1)水平的影响。方法选取C57/BL6小鼠48只,共分为6组,每组8只,通过高脂饮食建立肥胖模型小鼠,再经咪喹莫特(IMQ)乳膏诱导制成肥胖合并银屑病小鼠模型,分别给予凉血消银饮组及复方凉血消银组中药灌胃处理28 d。苏木精-伊红(HE)染色分析皮损情况,免疫组化法、酶联免疫吸附测定(ELISA)法分析复方凉血消银饮对肥胖合并银屑病模型小鼠代谢指标IGF-1、胰岛素样生长因子-1受体(IGF-1R)、胰岛素样生长因子结合蛋白-3(IGFBP-3)和角蛋白17(KPT17)的影响。结果复方凉血消银组模型小鼠体质量显著低于其他4组,和正常空白对照组相当;与模型组比较,复方凉血消银饮可明显降低银屑病合并肥胖模型小鼠银屑病面积和严重程度指数(PASI)评分,减轻其鳞屑红斑症状,差异有统计学意义(P<0.05);复方凉血消银饮干预后IGF-1、IGF-1R和IGFBP-3表达均与模型组比较明显降低,差异有统计学意义(P<0.05)。结论复方凉血消银饮可通过降低IGF-1、IGF-1R和IGFBP-3等细胞因子的表达有效抑制银屑病鳞屑产生及皮肤红肿、过度增生,同时有效降低体质量。Objective In order to explore the effects of the compound Liangxue Xiaoyin decoction on the levels of insulinlike growth factor-1(IGF-1)in mice models of psoriasis combined with obesity.Methods Fourty-eight C57/BL6 mice were selected and divided into six groups with eight mice in each group.An obesity model was established through a high-fat diet,followed by the induction of a psoriasis-like model using imiquimod(IMQ)ointment.The groups received the Liangxue Xiaoyin decoction and the compound Liangxue Xiaoyin decoction via gastric administration for 28 days.Hematoxylin and eosin(HE)staining was conducted to analyze skin lesions,and immunohistochemical methods along with enzyme-linked immunosorbent assay(ELISA)were utilized to assess the impact of the compound Liangxue Xiaoyin decoction on metabolic indicators IGF-1,insulin-like growth factor-1 receptor(IGF-1R),insulin-like growth factor binding protein-3(IGFBP-3),and keratin 17(KPT17)in the psoriasis and obesity model mice.Results Mice in the compound Liangxue Xiaoyin group exhibited a significant reduction in body weight compared to the other four groups,similar to the normal control group.The compound Liangxue Xiaoyin decoction markedly decreased the psoriasis area and severity index(PASI)scores of the psoriasis combined with obesity model mice,alleviating their erythematous scaling symptoms(P<0.05).Following intervention with the compound Liangxue Xiaoyin decoction,the expressions of IGF-1,IGF-1R,and IGFBP-3 were significantly reduced compared to the model group(P<0.05).Conclusion The compound Liangxue Xiaoyin decoction effectively inhibits the production of psoriatic scales and skin inflammation by reducing the expression of cytokines such as IGF-1,IGF-1R and IGFBP-3,while also significantly lowering body weight.
