摘 要:目的:探讨脐带间充质干细胞(Umbilical Cord Mesenchymal Stem Cells,UC-MSCs)对SD雌性大鼠宫腔粘连子宫内膜的修复作用。方法:选取18只SD雌性大鼠作为实验对象,随机均分为三组,各组含6只。首先确认大鼠的动情周期,随后进行如下操作:正常组大鼠仅行剖腹并缝合,不接受任何治疗,于术后7天处死;模型组大鼠剖腹后,使用眼科钳对子宫壁进行多次刮擦以建立宫腔粘连模型,7天后处死;UC-MSCs治疗组则在建模后7天再次开腹,向双侧子宫宫腔内灌注UC-MSCs,连续治疗三个动情周期后处死大鼠。收集所有大鼠的双侧子宫组织,进行包埋切片,并依次进行HE染色、MASSON染色及CK免疫组织化学染色。通过测量子宫内膜厚度、统计腺体数量、计算子宫内膜纤维化面积以及评估CK染色的平均光密度,来综合评价UC-MSCs在治疗宫腔粘连中的效果。结果:LSD-t检验和Dunn’st检验分析:相较于模型组,正常组与UC-MSCs治疗组的HE染色结果显示,子宫内膜厚度显著增加(F=86.361,P<0.001),腺体数目也显著增长(F=81.639,P<0.001);在MASSON染色中,与模型组对比,UC-MSCs治疗组显示出纤维化程度的显著降低(F=50.151,P<0.001);此外,正常组与UC-MSCs治疗组的CK染色平均光密度相较于模型组均有明显提升(F=10.326,P=0.002)结论:UC-MSCs可增加子宫内膜的厚度和腺体的数量,降低子宫内膜纤维化程度,提高角蛋白阳性表达率,促进子宫内膜组织修复,恢复其部分功能,起到治疗宫腔粘连的作用。Objective:To investigate the role of umbilical cord mesenchymal stem cells(UC-MSCs)in the repair of uterine adhesion endometrium in SD female rats.Methods:Eighteen SD female rats were selected as experimental subjects and randomly divided equally into three groups of six rats each.First,the estrous cycle of the rats was confirmed,and then the following operations were performed:In the model group,the rats were dissected and then the uterine wall was scraped several times with ophthalmic forceps to establish the model of uterine adhesion,and then the rats were sacrificed 7 days after surgery;In the UC-MSCs treated group,the rats were dissected again and the UC-MSCs were infused into the uterine cavity bilaterally,and the rats were treated for three consecutive cycles and then executed.The rats were killed after three consecutive cycles of treatment.Bilateral uterine tissues were collected from all rats,embedded in sections and subjected to HE staining,MASSON staining and CK immunohistochemical staining.The effect of UC-MSCs in the treatment of uterine adhesions was comprehensively evaluated by measuring endometrial thickness,counting the number of glands,calculating the area of endometrial fibrosis,and assessing the average optical density ofCK staining.To investigate the role of umbilical cord mesenchymal stem cells(UC-MSCs)in the repair ofuterine adhesion endometrium in SDfemale rats.Results:Compared with the model group,the HE staining results of the normal group and the UC-MSCs treated group showed a significant increase in endometrial thickness(F=86.361,P<0.001),while the number of glands also showed a significant increase(F=81.639,P<0.001).In MASSON staining,the UC-MSCs treated group showed a significant reduction in fibrosis compared to the control group(F=50.151,P<0.001).In addition,the mean optical density of CK staining was significantly increased in both normal and UC-MSCs treated groups compared to the model group(F=10.326,P=0.002).Conclusion:This study demonstrated that UC-MSCs increased endometrial t
