摘 要:【目的】改变文冠果传统的育种方式,对文冠果组织培养快速繁殖技术进行了探索,为文冠果组织培养、工厂化育苗、良种繁育提供理论参考。【方法】试验以优良品种文冠果幼嫩叶片作为外植体进行无菌培养,探索培养基中添加不同激素(6-BA、IAA、IBA)浓度组合对愈伤组织诱导、不定芽增殖及不定根诱导的影响。【结果】MS+6-BA 0.4 mg/L+IBA 1.5 mg/L+NAA 0.2 mg/L最适于文冠果叶片愈伤组织诱导,MS+6-BA 0.2 mg/L+NAA 0.1 mg/L+IBA 0.5 mg/L更利于不定芽增殖,1/2 MS+NAA 0.1 mg/L+IBA 2.0 mg/L对生根有促进作用。【结论】影响文冠果快速繁殖的激素种类较多,其中激素6-BA、IAA、IBA对文冠果生长影响较大,合适的培养基配以合理的激素浓度更有利于文冠果快速繁殖,为文冠果大规模生产提供了理论参考和科技支撑。【Objective】The research aimed to change the traditional breeding method of Xanthoceras sorbifolia Bunge,explore the rapid propagation technology of Xanthoceras sorbifolia Bunge tissue culture,and provide theoretical basis for Xanthoceras sorbifolia Bunge tissue culture,factory seedling cultivation and improved cultivar breeding.【Methods】In this experiment,the young leaves of Xanthoceras sorbifolia Bunge were used as explants for aseptic culture,and the effects of adding different hormone(6-BA,IAA,IBA)concentration combinations on callus induction,adventitious shoot proliferation and adventitious root induction were explored.【Results】The experimental results showed that MS+6-BA 0.4 mg/L+IBA 1.5 mg/L+NAA 0.2 mg/L were most suitable for inducing leaf callus tissue.MS+6-BA 0.2 mg/L+NAA 0.1 mg/L+IBA 0.5 mg/L were more conducive to the proliferation of adventitious buds.1/2 MS+NAA 0.1 mg/L+IBA 2.0 mg/L had a promoting effect on rooting.【Conclusion】There are many types of hormones that affect the rapid reproduction of Xanthoceras sorbifolium Bunge,among which hormones 6-BA,IAA,and IBA have a greater impact on the growth of Xanthoceras sorbifolium Bunge.Suitable culture media with reasonable hormone concentrations are more conducive to the rapid reproduction of Xanthoceras sorbifolium Bunge.This study provides a theoretical basis and technological support for the large-scale production of Xanthoceras sorbifolium Bunge.
