摘 要:目的利用微弧氧化(MAO)技术合成微弧氧化表面改性钛铜合金[Ti-10Cu(MAO)]并探究其对骨形成的影响及机制。方法利用MAO技术合成Ti-10Cu(MAO),MTT、细胞克隆实验检测细胞毒性及增殖能力;细胞划痕实验检测细胞迁移能力;碱性磷酸酶(ALP)染色和茜素红(ARS)染色评价MG63细胞分化及骨形成能力;Western blot检测OPN、Runx2蛋白表达情况;免疫荧光、Western blot检测血管内皮细胞(HUVEC细胞)VEGFR蛋白表达情况。结果与对照组相比,Ti、Ti-10Cu和Ti-10Cu(MAO)对MG63细胞增殖能力无显著抑制作用,细胞划痕间伤口愈合距离百分比与对照组之间没有明显差异,说明上述材料安全无毒性;ALP与ARS染色结果显示Ti-10Cu(MAO)可显著增加ALP与ARS阳性表达,并且显著促进成骨相关蛋白OPN,Runx2蛋白表达;此外,Ti-10Cu(MAO)可显著促进HUVEC细胞VEGFR蛋白的表达。结论微弧氧化表面改性处理后的Ti-10Cu合金,具有较好的生物安全性,能够促进骨形成,其作用机制是通过上调VEGFR蛋白表达,促进血管再生,从而进一步促进成骨。Objective Micro-arc oxidation(MAO)technology was used to synthesize surface modified titanium-copper alloy[Ti-10Cu(MAO)]and explored its effect on bone formation and mechanism.Methods Ti-10Cu(MAO)was synthesized using MAO technology.Cytotoxicity and proliferation ability were detected by MTT and cell cloning assay.Cell migration ability was detected by cell wound healing.Alkaline phosphatase(ALP)and alizarin red(ARS)staining were used to evaluate the differentiation and bone formation ability of MG63 cells.The expressions of OPN and Runx2 were detected by Western blot.The expression of VEGFR in vascular endothelial cells was detected by immunofluorescence and Western blot.Results Compared with control group,Ti,Ti-10Cu and Ti-10Cu(MAO)had no significant inhibitory effect on the proliferation ability of MG63 cells,and the wound healing rate of the cells was not significantly different from that of the control group,indicating that the above materials were safe and non-toxic.The results of ALP and ARS staining showed that Ti-10Cu(MAO)significantly increased the positive expression of ALP and ARS,and significantly promoted the expressions of OPN and Runx2 proteins related to osteogenesis.In addition,Ti-10Cu(MAO)significantly promoted the expression of VEGFR protein in HUVEC cells.Conclusion Ti-10Cu after surface modification by micro-arc oxidation has good biosafety and can promote bone formation.The mechanism of action may be to promote vascular regeneration through up-regulation of VEGFR protein expression,thus further promoting bone formation.
