摘 要:目的探究重组人生长激素(r-hGH)对大鼠烧伤创面愈合的影响及相关机制。方法将SD大鼠随机分为模型组和药物处理组,每组10只。药物处理14 d后,大体观察大鼠皮肤创面愈合情况;HE染色检测大鼠皮肤创面组织病理学情况;ELISA检测大鼠血清IgG、IgA和IgM表达水平;流式细胞术检测大鼠血清CD4^(+)和CD8^(+)T细胞亚群水平,并计算CD4^(+)/CD8^(+)数值;Western blot检测大鼠皮肤创面组织JAK2/STAT3信号通路蛋白表达情况。结果r-hGH处理增加大鼠烧伤皮肤创面愈合速度;上调大鼠血清IgG、IgA和IgM表达水平、CD4^(+)水平和CD4^(+)/CD8^(+)数值,下调大鼠血清CD8^(+)水平;降低大鼠烧伤皮肤创面组织p-JAK2和p-STAT3蛋白表达水平。结论r-hGH改善烧伤大鼠免疫功能并促进创面愈合,其机制与抑制JAK2/STAT3信号通路激活有关。Objective To explore the effect of recombinant human growth hormone(r-hGH)on burn wound healing in rats and its related mechanism.Methods SD rats were randomly divided into model group and drug treatment group with 10 rats in each group.After 14 days of drug treatment,the skin wound healing of rats was observed.The histopathology of skin wounds of rats was detected by HE staining.The levels of IgG,IgA and IgM in serum of rats were detected by ELISA.CD4^(+)and CD8^(+)T cell subsets in serum were detected by flow cytometry,and CD4^(+)/CD8^(+)values were calculated.The expressions of JAK2/STAT3 signaling pathway in skin wound tissue of rats were detected by Western blot.Results Treatment with r-hGH increased the healing rate of burned skin wounds in rats,up-regulated the levels of IgG,IgA and IgM,CD4^(+)and CD4^(+)/CD8^(+),and down-regulated the level of CD8^(+)in serum,reduce the expressions of p-JAK2 and p-STAT3 proteins in burnt skin wound tissue of rats.Conclusions r-hGH can improve immune function and promote wound healing in burn rats,and its mechanism may be related to inhibiting the activation of JAK2/STAT3 signaling pathway.
