摘 要:目的探讨低温环境对大鼠甲状腺放射性损伤状态的影响。方法60只SD大鼠随机分为正常对照组(NC组)、低温无辐射组(-10℃,LT_(1)组)、极低温无辐射组(-20℃,LT_(2)组)、常温辐射组(RT+R组)、低温辐射组(-10℃,LT_(1)+R组)和极低温辐射组(-20℃,LT_(2)+R组),每组10只。采用^(131)I-NaI溶液灌胃的方式制造大鼠甲状腺放射性损伤模型,ELISA方法检测损伤后4周大鼠血清FT_(3)、FT_(4)、TSH、TgAb水平;苏木素伊红(HE)染色和TUNEL检测大鼠甲状腺组织细胞损伤状态和凋亡水平。Western blot方法检测大鼠甲状腺组织组织中XIAP和caspase3蛋白表达。结果在放射性照射后,低温环境中大鼠血清的FT_(3)、FT_(4)水平有所减低,TSH水平升高,甲状腺组织XIAP表达升高,caspase3表达降低;极低温环境下,FT_(3)、FT_(4)减低及TSF升高程度进一步加重;同等辐射剂量下,在低温环境中的大鼠甲状腺细胞凋亡程度较常温时减轻;在极低温环境中,甲状腺细胞凋亡率进一步上升。结论低温环境对SD大鼠的甲状腺放射性损伤具有一定的保护作用,而在极低温度下,甲状腺组织被进一步破坏,损伤加重。Objective To investigate the effect of cold environment on the status of thyroid radiation damage in rats.Methods Sixty SD rats were randomly divided into normal control group(NC group),low temperature without radiation group(-10℃,LT 1),very low temperature without radiation group(-20℃,LT 2),normal temperature radiation group(RT+R group),low temperature radiation group(-10℃,LT 1+R),and very cold radiation group(-20℃,LT 2+R),10 per group.The rat thyroid radiation damage model was created by^(131)I-NaI solution,and the levels of FT3,FT4,TSH,TgAb in serum were measured by ELISA at 4 weeks after injury.Hematoxylin eosin(HE)staining and TUNEL were used to detect the injury status and apoptosis of cells in thyroid tissue of rats.The expressions of XIAP and caspase3 in thyroid tissue of rats were detected by Western blot.Results After radiation exposure,the levels of FT_(3) and FT_(4) in serum of rats were decreased,level of TSH was increased,the expression of XIAP in thyroid tissue of rats was increased and the expression of caspase3 was decreased.At extremely low temperature,the levels of FT_(3) and FT_(4) were decreased and the level of TSF was increased further.Under the same radiation dose,the apoptosis degree of thyroid cells in low temperature environment was less than that in normal temperature environment.In the extremely low temperature environment,the apoptosis rate of thyroid cells was increased further.Conclusion Cold environment has some protective effect on thyroid radiation damage in SD rats,while at very low temperature,thyroid tissue is further destroyed and the damage is aggravated.
