摘 要:目的基于cAMP/PKA信号通路探讨养心汤治疗室性心律失常的作用机制。方法将40只雄性SD大鼠随机分为空白组10只、模型组30只。模型组大鼠尾静脉注射乌头碱,建立室性心律失常模型。将造模成功大鼠根据随机数字表法分为模型对照组、胺碘酮组和养心汤组,每组10只。利用生物信号采集及分析系统描记各组大鼠肢体标准Ⅱ导联心电图;ELISA测定各组大鼠血浆cAMP、PKA浓度;流式细胞仪测定各组大鼠Ca^(2+)浓度;Western blot检测各组大鼠心肌组织PKA、RyR2、Gi蛋白表达水平。结果养心汤可以延长室性心律失常模型大鼠VA出现时间,缩短VA持续时间;升高室性心律失常模型大鼠血浆中cAMP及PKA含量,降低心肌细胞中Ca^(2+)浓度;降低室性心律失常模型大鼠心肌组织中PKA及RyR2蛋白表达水平,升高Gi蛋白表达水平。结论养心汤可通过调控cAMP/PKA信号通路对室性心律失常模型大鼠起治疗作用。Objective To investigate the therapeutic mechanism of Yangxin Decoction for ventricular arrhythmia based on the cAMP/PKA signaling pathway.Methods Fortymale SD rats were randomly divided into blank group(10 rats)and model group(30 rats).The model group was injected with aconitine via the tail vein to establish a ventricular arrhythmia model.Successfully modeled rats were then randomly assigned to the model control group,amiodarone group,and Yangxin Decoction(YX)group,with 10 rats in each group.A biological signal acquisition and analysis system was used to record the standard limb lead Ⅱ electrocardiogram of the rats in each group.ELISA was used to measure the plasma cAMP and PKA concentrations of rats in each group.Flow cytometry was employed to determine the Ca^(2+)concentration in the myocardial cells of rats in each group.Western blot was used to detect the expressions of PKA,RyR2,and Gi proteins in the myocardial tissues of rats in each group.Results Yangxin Decoction can prolong the time of VA occurrence and shorten the duration of VA in rats with ventricular arrhythmia.The concentration of cAMP and PKA in plasma of ventricular arrhythmia model rats was increased,and Ca^(2+)concentration in cardiomyocytes was decreased.The expressions of PKA and RyR2 protein in ventricular arrhythmia model rats were decreased,and the expression of Gi protein was increased.Conclusions Yangxin Decoction has a therapeutic effect on ventricular arrhythmia model rats by regulating cAMP/PKA signaling pathway.
