摘 要:目的探讨柴胡皂苷A对嗅球切除小鼠抑郁样行为及海马环磷酸腺苷反应元件结合蛋白(CREB)表达的影响。方法30只SPF级雄性C57BL/6J小鼠,6周龄,采用随机数字表法分为3组(每组n=10):对照组、嗅球切除组、柴胡皂苷A组。采用嗅球切除术损毁嗅球切除组及柴胡皂苷A组小鼠的嗅球,而对照组小鼠仅在相同坐标进行颅骨钻孔,不毁损嗅球。通过灌胃途径给予柴胡皂苷A组小鼠柴胡皂苷A溶液(剂量:5 mg/kg),对照组和嗅球切除组小鼠给予相同体积0.9%NaCl溶液,1次/d,持续21 d。采用旷场实验、悬尾实验、强迫游泳实验及体质量称量评估各组小鼠抑郁相关的行为学变化。采用Western blot检测各组小鼠海马CREB的表达。采用Graphpad prism 5进行数据分析,多组间数据比较采用单因素方差分析,进一步两两比较采用Tukey检验。结果(1)体质量测定结果显示:嗅球切除术当天,各组小鼠体质量差异无统计学意义(F=0.468,P=0.631),嗅球切除28 d,3组小鼠体质量差异有统计学意义(F=37.340,P<0.001)。嗅球切除组小鼠体质量低于对照组[(24.09±1.01)g,(27.22±0.58)g](P<0.05)。柴胡皂苷A组小鼠体质量[(26.23±0.83)g]高于嗅球切除组(P<0.05)。3组小鼠体质量差值差异有统计学意义(F=48.180,P<0.001)。嗅球切除组小鼠体质量差值低于对照组[(2.34±0.96)g,(5.57±0.72)g](P<0.05)。柴胡皂苷A组小鼠体质量差值高于嗅球切除组[(4.74±0.56)g,(2.34±0.96)g](P<0.05)。(2)旷场实验结果显示,3组小鼠旷场中心停留时间和运动总路程均差异有统计学意义(F=27.03,10.98,均P<0.001)。嗅球切除组小鼠旷场中心停留时间[(13.87±4.59)s]和运动总路程[(35.67±4.88)m]均短于对照组[(27.44±2.76)s,(59.02±16.21)m](均P<0.05)。柴胡皂苷A组小鼠旷场中心停留时间[(21.77±4.78)s]和运动总路程[(52.33±10.41)m]均长于嗅球切除组(均P<0.05)。(3)悬尾实验结果显示,3组小鼠不动时间差异有统计学意义(F=74.57,P<ObjectiveTo investigate the effect of saikosaponin A on depressive-like behaviors and hippocampal cyclic adenosine monophosphate response element-binding(CREB)protein expression in olfactory bulbectomized mice.MethodsTotally thirty male SPF-grade C57BL/6J mice,six weeks old,were randomly divided into 3 groups(n=10 in each group):the control group,the olfactory bulbectomy group,and the saikosaponin A group.Olfactory bulbectomy was performed to damage the olfactory bulbs in the mice of bulbectomy group and saikosaponin A group,while the mice of control group only had a cranial drill hole at the same coordinates without damaging the olfactory bulbs.Mice in saikosaponin A group were administered saikosaponin A solution(dose:5 mg/kg)via gavage,while mice in the control and olfactory bulbectomy groups were given the same volume of 0.9%NaCl solution once a day for 21 days.The depressive-related behavioral changes of mice in each group were evaluated by open field test,tail suspension test,forced swimming test,and weight measurement.The expression of CREB in the hippocampus of mice were detected by Western blot.Graphpad prism 5 software was used for data analysis.The data comparison among the multiple groups was conducted by one-way ANOVA and further pairwise comparisons were conducted by Tukey test.Results(1)The results of weight measurement showed that there was no significant difference in body weight among the three groups(F=0.468,P=0.631)on the day of the olfactory bulbectomy surgery,but there was a statistically significant difference in body weight among the three groups(F=37.340,P<0.001)on the 28th day after the olfactory bulbectomy.The body weight of mice in the olfactory bulbectomy group was lower than that of the control group((24.09±1.01)g,(27.22±0.58)g)(P<0.05).The body weight of mice in the saikosaponin A group((26.23±0.83)g)was higher than that of the olfactory bulbectomy group(P<0.05).There was a statistically significant difference in body weight differences among the three groups of mice(F=48.180,P<0
