摘 要:旨在探究刚地弓形虫表面抗原相关序列蛋白SRS40E的结构功能以及其作为抗弓形虫疫苗的候选分子的潜力。通过NCBI数据库下载表面抗原相关序列蛋白SRS40E的基因序列和氨基酸序列,通过ORFfinder网站联合GENSCAN网站对其基因序列进行ORF分析,氨基酸序列依次通过Prot Param、Expasy-ProtScale、TMHMM、SignalP-4.1、SOPMA、SWISS-MODEL、IEDB、SYFPEITHI、VaxiJen 2.0程序对SRS40E蛋白的理化性质、亲疏水性、跨膜结构、信号肽、空间结构、磷酸化位点、抗原表位进行分析预测。结果:表面抗原相关序列蛋白SRS40E的基因序列全长1 627 bp,由396个氨基酸组成,理论分子量是42 204.65,等电点为7.87,分子式为C_(1818)H_(2951)N_(513)O_(600)S_(19),不稳定指数为47.19,脂溶性指数为72.73,亲水性总平均值为-0.314,预测显示为亲水性蛋白;该蛋白没有信号肽,具有跨膜结构;在二级结构中,α-螺旋占比18.94%、β-转角占比2.53%、β-折叠占比20.71%、无规则卷曲占比为57.83%;有53个磷酸化位点,其中30个丝氨酸磷酸化位点,22个苏氨酸磷酸化位点,1个酪氨酸磷酸化位点;有18个抗原决定簇,8个优势B细胞表位,8个优势辅助性T细胞(Th)细胞表位和2个优势细胞毒性T淋巴细胞(CTL)细胞表位。综上,表面抗原相关序列蛋白SRS40E包含多个B、T细胞表位,可作为弓形虫疫苗的候选蛋白。This study was to investigate the structural and functional characteristics of the surface antigen SAG1-related sequence protein SRS40E of Toxoplasma gondii and its potential as a candidate molecule for anti-Toxoplasma vaccines.The gene and amino acid sequences of the surface antigen SAG1-related sequence protein SRS40E were downloaded from the NCBI database.The gene sequence was analyzed by ORFfinder website in conjunction with GENSCAN.The amino acid sequence was then subjected to analyze using Prot Param,Expasy-ProtScale,TMHMM,SignalP-4.1,SOPMA,SWISS-MODEL,IEDB,SYFPEITHI,and VaxiJen 2.0 programs to predict and analyze the physicochemical properties,hydrophilicity,transmembrane structure,signal peptide,spatial structure,phosphorylation sites and antigenic epitopes of surface antigen SAG1-related sequence protein SRS40E.The results showed that the surface antigen SAG1-related sequence protein SRS40E possessed a total length of 1627 bp and consisted of 396 amino acids with a theoretical molecular weight of 42204.65,an isoelectric point of 7.87 and a molecular formula of C_(1818)H_(2951)N_(513)O_(600)S_(19).It exhibited an instability index of 47.19,a lipid solubility index of 72.73,and an overall hydrophilicity average of-0.314,indicating it was a hydrophilic protein.The protein lacked a signal peptide but possessed a transmembrane structure.In its secondary structures,α-helices accounted for 18.94%,β-turns for 2.53%,β-strands for 20. 71%,and irregular coils for 57. 83%. There were 53 phosphorylation sites,including 30 serine phosphorylation sites,22 threonine phosphorylationsites,and 1 tyrosine phosphorylation site. Additionally,there were 18 antigenic determinant clusters,8 dominant B - cellepitopes,8 dominant Th-cell epitopes and 2 dominant CTL-cell epitopes. In conclusion,the surface antigen SAG1-related sequence proteinSRS40E contained multiple B and T cell epitopes,being qualified as a candidate protein for a Toxoplasma vaccine.
