一例鹅星状病毒2型感染的实验室诊断与分析

摘　　要：为查明山东省部分朗德鹅养殖场填食鹅发病死亡的原因,本研究取6只病死鹅,剖检观察大体病变,采集明显病变组织,利用PCR方法进行禽流感病毒、新城疫病毒、鹅星状病毒2型(GoAstV-2)、坦布苏病毒、禽呼肠孤病毒、鹅细小病毒、传染性法氏囊病毒、传染性支气管炎病毒、禽腺病毒的核酸检测,利用鹅胚进行病毒分离,并对病毒ORF1b基因全长测序。结果:剖检显示死鹅肾脏肿大,肠黏膜、泄殖腔出血,内脏尿酸盐沉积;结合临床症状、剖检结果、PCR鉴定结果、遗传进化及同源性分析结果,确认该病例为GoAstV-2感染;同时,将病毒分离株感染鹅进行动物致病性试验,剖检可见感染鹅各组织器官有明显的尿酸盐沉积,肝脏、肾脏出现病理组织损伤。本文通过对一例GoAstV-2感染的实验室诊断进行分析,为GoAstV-2型的进一步研究提供参考。This study was to determine the causes of morbidity and mortality of feeding geese in some Landaise goose farms in Shandong Province.Six dead geese were selected.Their gross lesions were observed and the obvious lesions were collected.Then,nucleic acid detection of avian influenza virus,Newcastle disease virus,goose astrovirus type 2,Tambusu virus,goose parvovirus,infectious bursal disease virus,infectious bronchitis virus,Avian arc enterovirus and Avian adenovirus were performed using PCR.The viruses were isolated and sequenced using goose embryos.And the ORF1b gene was sequenced.The autopsy results showed that the dead geese had kidney enlargement,intestinal mucosa,cloaca bleeding,viscera urate deposition.Combined with the clinical symptoms,the PCR identification results,the genetic evolution and homology analysis results,the case was confirmed as goose astrovirus type 2 infection.At the same time,the geese were infected with the virus isolate for animal pathogenicity test,and Necropsy showed that the infected geese had obvious urate deposition in various tissues and organs,and pathological tissue damage in their liver and kidney.In this paper,the laboratory diagnosis of goose astrovirus type 2 infection was analyzed,which provided reference for further research on goose astrovirus type 2.
