摘  要:为研究苹果溶质转运蛋白家族基因MdSLC25A44-like的序列和表达模式,利用荧光定量及生物信息学技术分析该基因序列特征及在不同类型苹果组织中的表达水平,分析结果显示,MdSLC25A44-like蛋白序列与白梨PbSLC25A44-like同源性最高;通过构建系统进化树得出,MdSLC25A44-like与白梨、秋子梨×西洋梨亲缘关系最近;生物信息学分析结果表示MdSLC25A44-like蛋白是一个α螺旋转运蛋白、稳定性蛋白并具有疏水性,定位于线粒体;花青苷测定结果表示,苹果红色果皮和红色果肉中的花青苷含量明显高于绿色果皮和白色果肉;实时荧光定量PCR结果表明,在苹果红色果皮和红色果肉中MdSLC25A44-like基因的表达量显著高于绿色果皮和白色果肉。MdSLC25A44-like基因可能参与了花青苷积累过程,基因的表达与花青苷积累量呈正相关关系,本结果为进一步研究苹果果实花青苷积累与转运以及提高果实果色品质提供了一定的科学基础。In order to study the sequence and expression pattern of the apple solute transport protein family gene MdSLC25A44-like in Malus domestica,the sequence characteristics and tissue expression patterns in the peel and pulp of different cultivars are analyzed using qRT-PCR and bioinformatics analysis.The results show that MdSLC25A44-like protein has the highest homology with PbSLC25A44-like from white pear.Phylogenetic analysis shows it has the closest affinity to white pear and autumn pear×Western pears.The bioinformatics analysis results indicate that the MdSLC25A44-like protein is anα-Spiral transporter protein,stable protein,and hydrophobic,located in mitochondria.The results of anthocyanin determination show that the content of anthocyanins in the red skin and red flesh of apples is higher than that in the green skin and white flesh.The real-time fluorescence quantitative PCR results show that the expression level of MdSLC25A44-like gene is higher in the red skin and red flesh of apples than in the green skin and white flesh.The above results indicate that the MdSLC25A44-like gene may be involved in the process of anthocyanin accumulation,and the expression of the gene is positively correlated with the amount of anthocyanin accumulation.This result provides a scientific basis for further research on the accumulation and transportation of anthocyanins in apple fruits and for improving fruit color quality.
