摘  要:目的探讨不同浓度一氧化氮(nitric oxide,NO)对胰岛β细胞活性和胰岛素分泌量的影响并探明NO积极作用的浓度范围。方法对体外培养的小鼠胰岛细胞株(Min6)进行分组,实验组以硝普钠(sodium nitroprusside,SNP)为NO供体,分别用0.1、1、2、4、6、8、10、20、40、60 mmol/L的SNP处理,各组采用硝酸还原酶法测定NO含量、CCK8法检测细胞活性、酶联免疫吸附试验(ELISA)检测胰岛素含量。结果SNP在0.1~60 mmol/L范围与其NO释放量呈线性关系(r=0.9666),且NO在2 h内稳定(P>0.05);NO在5.38μmol/L时Min6细胞活力与对照组比较,差异无统计学意义(P>0.05);在5.38~302.21μmol/L时细胞活力随NO浓度增加而降低(r=-0.9959,P<0.05),>302.21μmol/L时细胞活力<5%;胰岛素随NO浓度增加呈先升高后降低趋势,NO在5.38~52.04μmol/L时胰岛素逐渐升高(52.04μmol/L时显著升高,P<0.05),>52.04μmol/L时胰岛素呈下降趋势(1182.11μmol/L时显著降低,P<0.05);在NO作用下,细胞活性与胰岛素分泌量总体呈正相关性(P<0.05),但NO<52.04μmol/L时胰岛素分泌量随细胞活力降低反而增大。结论小鼠胰岛β细胞活性及胰岛素分泌量与NO浓度总体呈正相关,但二者在不同NO浓度范围的变化趋势不同,NO在5.38~52.04μmol/L时浓度增加对胰岛素分泌有促进作用。Objective To explore the effects of different concentrations of nitric oxide(nitric oxide,NO)on pancreaticβcell activity and insulin secretion,and to determine the concentration range of positive effect of NO.Methods The mouse isletβcell line(Min6)was divided into groups for in vitro culture.In the experimental group,sodium nitroprusside(sodium nitroprusside,SNP)was used as NO donor,and SNP was treated with 0.1,1,2,4,6,8,10,20,40,and 60 mmol/L respectively.The nitrate reductase method was used to determine the NO content in each group.Cell activity was detected by CCK8 method and insulin content was detected by enzyme-linked immunosorbent assay(ELISA).Results The SNP showed a linear relationship with NO release in the range of 0.1-60 mmol/L(r=-0.9666),and NO was stable within 2 h(P>0.05).NO at 5.38μmol/L,the cell viability of Min6 was not statistically significant,compared with the control group(P>0.05).Cell viability decreased with increasing NO concentration at 5.38-302.21μmol/L(r=-0.9959,P<0.05).When greater than 302.21μmol/L,the cell viability was less than 5%.Insulin increased with the increase of NO concentration and then decreased.NO gradually increased with insulin at 5.38-52.04μmol/L(significantly increased at 52.04μmol/L,P<0.05).Insulin showed a downward trend when greater than 52.04μmol/L(significantly decreased at 1,182.11μmol/L,P<0.05).Under the action of NO,cell activity was positively correlated with insulin secretion(P<0.05).However,when NO<52.04μmol/L,insulin secretion increased with the decrease of cell viability.Conclusion The activity ofβcells and insulin secretion in mouse islets are positively correlated with NO concentration,but the trend of change is different in different NO concentration ranges.NO increases the concentration at 5.38-52.04μmol/L,which promotes insulin secretion.
