摘 要:探究XsbZIP17对文冠果抗旱性的影响,以增加对文冠果抗旱性调控网络的认识,选育抗旱性更强的文冠果品种。本研究利用生物信息学方法分析XsbZIP17基因的结构特征,构建35S∷XsbZIP17-eYFP融合蛋白,对XsbZIP17进行亚细胞定位,并通过实时荧光定量PCR方法明确XsbZIP17在文冠果根、茎、嫩叶、老叶和花5个组织部位的表达情况;将构建的XsbZIP17基因过表达载体异源转化到拟南芥,并观察干旱胁迫下野生型植株和过表达转基因株系的生长表型,测定脯氨酸、丙二醛和叶绿素指标以及其他抗旱性相关基因的表达水平。研究发现,XsbZIP17具有bZIP家族保守结构域bZIP-HY5-like,并定位于细胞核中。XsbZIP17在文冠果根和茎中表达量较高,并且其表达受到干旱信号的强烈诱导。在干旱胁迫下,XsbZIP17过表达株系的存活率显著高于野生型,其脯氨酸含量和叶绿素含量也高于野生型,而丙二醛含量低于野生型;并且在过表达拟南芥中,抗旱性相关基因的表达水平均显著上升。综上所述,XsbZIP17在调控文冠果抗旱性中发挥了重要的作用;在干旱条件下,XsbZIP17能提高转基因拟南芥的脯氨酸含量,以维持植物的渗透压平衡,减轻植物质膜过氧化程度,还可以提高叶绿素含量,以维持植物正常的光合作用,从而提高植株抗旱性。The objective of this study was to explore the effect of XsbZIP17 on the drought resistance of yellowhorn(Xanthoceras sorbifolium)for increasing the understanding of the drought resistance regulatory network of yellowhorn,and for selecting yellowhorn varieties with stronger drought resistance.Bioinformatics methods was adopted to analyze the structural features of XsbZIP17 gene,and the 35S∷XsbZIP17-eYFP fusion protein was constructed to observe the subcellular localization of XsbZIP17.The expression pattern of XsbZIP17 in five tissues of yellowhorn,including roots,stems,young leaves,mature leaves,and flowers,were clarified through qRT-PCR.XsbZIP17 was heterologously overexpressed by Agrobacteriummediated genetic technology to obtain transgenic Arabidopsis thaliana.We observed the growth phenotypes of wild type(WT)and transgenic plants under drought stress,measured their physiological indicators of proline,malondialdehyde,and chlorophyll,and expression levels of drought resistance related genes.The research has found that XsbZIP17 possesses the conserved structural domain bZIP-HY5-like characteristic of the bZIP family and is located in the nucleus.XsbZIP17 had a higher expression level in the stems and roots of yellowhorn,and its expression was strongly induced by drought and PEG treatment.Under drought stress,the survival rate of XsbZIP17 overexpression(XsbZIP17-OE)lines was significantly higher than that of the WT,and the proline and chlorophyll contents of XsbZIP17-OE lines were also higher than that of the WT,while the MDA content of XsbZIP17-OE lines was lower than that of the WT.And the expression levels of genes related to drought response in transgenic plants significantly increased.In summary,XsbZIP17 plays an important role in regulating the drought resistance of yellowhorn.XsbZIP17 increases the proline content of transgenic plants under drought conditions to maintain osmotic pressure balance,reduces the degree of plant plasma membrane peroxidation,and increases the chlorophyll content to maintain no
