摘  要:以红桦(Betula albosinensis)花粉为材料,探究其离体培养萌发的最适培养基和培养温度,比较了TTC、I-KI染色法及醋酸洋红法测定红桦花粉活力的准确性;同时从花粉含水量、容器是否抽取真空以及贮藏温度等方面探讨其贮藏效果。结果表明,红桦花粉离体培养的适宜培养基为10 g/L琼脂+15%蔗糖+250 mg/L硼酸,温度30℃,培养10 h活力最强;TTC染色法适宜在野外快速检测红桦花粉活力,且染色液pH为7.0时检测效果最好;红桦花粉经干燥和抽真空处理后有利于贮藏,最佳贮藏温度为-80~-40℃。本研究明确了红桦花粉特性及贮藏特性,为红桦杂交育种及种质创新提供了理论依据。Using Betula albosinensis pollen as a material,we explored the optimal medium for its in vitro culture germination,and compared the accuracy of TTC staining,I-KI staining,and magenta acetate method in determining the viability of the pollen;meanwhile,we discussed the storage effect of the pollen in terms of its water content,whether the container was vacuumed or not,and the storage temperature.The results showed that the suitable medium and culture temperature for in vitro culture of the pollen were 10 g/L agar+15%sucrose+250 mg/L boric acid and 30℃,10 h,the activity was the highest,and TTC staining was suitable for the rapid detection of the viability of the pollen in the field,and the best effect was achieved when the pH of the staining solution was 7.0.The optimal storage temperature of the pollen were-80 to-40℃,and B.albosinensis pollen was favorable for storage after dried and vacuumed.The study clarifies the characteristics and storage characteristics of B.albosinensis pollen,and provides a theoretical basis for the hybrid breeding and germplasm innovation of B.albosinensis.
