摘 要:研究旨在通过检测肌内脂肪(IMF)与腹脂(AF)差异沉积肉鸡的血清代谢组和脂质组,筛选影响肌内脂肪与腹脂差异沉积的关键代谢物。以快速型黄羽肉鸡广西金陵花鸡的终端父系E系为试验素材,对肌内脂肪与腹脂差异沉积极端个体的血清进行代谢组学和脂质组学研究,挖掘与之相关的血清关键代谢物。综合运用加权基因共表达网络分析、双向正交偏最小二乘法、受试者工作特征曲线和随机森林等方法,鉴定到鸡血清中20种体脂差异分布相关的关键候选物质。进一步的文昌鸡血清广靶代谢组验证结果表明,1-硬脂酰-2-花生四烯酰-sn-甘油-3-磷酸丝氨酸是肌内脂肪与腹脂差异沉积的关键代谢物。结果表明,1-硬脂酰-2-花生四烯酰-sn-甘油-3-磷酸丝氨酸,甘油磷脂代谢通路是影响肉鸡肌内脂肪与腹脂差异沉积的关键代谢物和关键通路。The aim of this study was to screen the key metabolites affecting the differential deposition of intramuscular fat(IMF)and abdominal fat(AF)by detecting the serum metabolomic and lipidomic of broiler chickens with differential deposition of IMF and AF.In this study,terminal sire line E of the fast-growing Guangxi Jinling yellow-feathered broilers was used as experimental material.Serum from individuals with extreme differences in IMF and AF deposition underwent metabolomic and lipidomic analyses to identify key serum metabolites associated with these differences.Using weighted gene co-expression network analysis,two-way orthogonal PLS,receiver operating characteristic curve and random forest methods,20 key candidate substances related to the differential distribution of body fat in chicken serum were identified.Further targeted metabolomic validation in Wenchang chicken serum revealed that 1-stearoyl-2-arachidonoyl-sn-glycerol-3-phosphoserine was a key metabolite for differential deposition between IMF and AF.The results indicated that 1-stearoyl-2-arachidonoyl-sn-glycerol-3-phosphoserine and the glycerophospholipid metabolism pathway were key metabolites and pathways that influenced the differential deposition of intramuscular fat and abdominal fat in broilers.
