摘 要:试验旨在探究芹黄素对鸡胚原代肝细胞氧化应激损伤的缓解作用。试验将鸡胚原代肝细胞(Chicken embryo liver,CEL)分为对照组、油酸组和芹黄素缓解组,每组5个重复,处理时间为12 h,处理结束后,进行油红O染色并收集细胞沉淀用于检测抗氧化相关指标。结果显示:油红O染色后,与对照组相比,油酸组的细胞可观察到明显的红色脂滴;且油酸组谷胱甘肽过氧化物酶(GPX)、谷胱甘肽S转移酶(GST)和超氧化物歧化酶(SOD)活性均显著降低(P<0.05),核因子E2相关因子2(Nrf2)、血红素加氧酶1(HO-1)、SOD和GST mRNA水平也显著下降(P<0.05);0.5~4μmol/L芹黄素对鸡胚原代肝细胞增殖无显著影响(P>0.05),5、6和10μmol/L芹黄素可显著降低鸡胚原代肝细胞的增殖活力(P<0.01)。此外,与油酸组相比,1μmol/L芹黄素处理后鸡胚原代肝细胞总抗氧化能力显著升高(P<0.05),而Nrf2、HO-1和GPX1 mRNA表达量只在数值上有增加,差异不显著(P>0.05)。综上所述,芹黄素对油酸所诱导的鸡胚原代肝细胞氧化应激具有缓解效果,但具体的作用机制仍需进一步研究。The study was aimed to investigate the effects of apigenin on oxidative stress injury of primary chicken embryo liver(CEL)cells were divided into the control group,oleic acid group and apigenin remission group with 5 replicates in each group,and the duration of the processing was 12 hours.After the treatment,oil red O staining was performed and cell precipitation was collected for the detection of antioxidant-related indicators.The result of oil red O staining showed that after the treatment of oleic acid,there was noticeable red lipid droplets in CEL cells;further,when compared with the control group,the activities of glutathione peroxidase(GPX),glutathione S transferase(GST)and superoxide dismutase(SOD)exhibited a significant reduction(P<0.05)and the mRNA levels of nuclear factor erythroid related factor 2(Nrf2),heme oxygenase(HO-1),SOD,and GST also showed a notable reduction(P<0.05).In the other hand,the CCK-8 results indicated that concentrations of apigenin ranging from 0.5 to 4μmol/L had no significant difference in hepatocyte viability,whereas the hepatocyte viability experienced a notable decrease at 5,6 and 10μmol/L apigenin(P<0.01).Additionally,when compared to the oleic acid group,the total antioxidant capacity(T-AOC)in the cell treatment with 1μmol/L apigenin increased significantly(P<0.05),while the expression levels of Nrf2,HO-1 and GPX1 were only numerically upregulated,with no significant difference(P>0.05).The results suggested that apigenin could alleviate oxidative stress induced by oleic acid,however,further investigation was needed to clarify the specific underlying mechanism.
