摘  要:为研究不同碳源稀释液配方对精液4℃保存过程中鸡精子活力的影响,试验以松阳锦鸡为研究对象,以果糖、蔗糖、葡萄糖和蜂蜜配备单碳源稀释液,评估不同碳源物质对精子活力的影响。同时,为进一步探究蔗糖与果糖适宜配比,按照蔗糖与果糖11个不同的比例配制稀释液,研究其对精子活力的影响。结果显示:果糖与蔗糖单碳源组各时间节点精子活力显著优于葡萄糖和蜂蜜单碳源组(P<0.05)。4℃存放1 h,果糖单碳源组精子活力显著高于其他组(P<0.05),4℃存放1.5 h和2 h,蔗糖作为单碳源组精子活力显著高于其他组(P<0.05);4℃存放0.5 h和1 h,果糖作为单碳源组精子前向运动比率显著高于其他组(P<0.05)。综上可得,使用蔗糖和果糖为碳源的稀释液进行鸡精液低温保存效果最好。随着4℃保存时间的延长,所有组别精子活力呈现显著下降(P<0.05)。B组稀释液在第5天精子前向运动比率显著高于其他组别(P<0.05),第10天的前向运动比率除C组外显著高于其他组(P<0.05),10 d的精子活力除C组外显著高于其他组(P<0.05)。随着蔗糖比例的增加,精子在4℃条件下前向运动活率降低明显。研究表明,通过合理的稀释液配方优化,配方B能够有效延长鸡精液体外保存时间,且能维持较好的精子运动形态,提高精子在低温状态下的存活时间,进而提高精子体外利用效率。Songyang Jin chickens was used as the research object,aiming to study the effects of different carbon source diluent formulas on the sperm vitality of chickens during the 4℃preservation process.The experiment firstly evaluated the effects of different carbon sources,including fructose,sucrose,glucose and honey on sperm vitality when used as sole carbon sources in the diluent.By preparing diluents with 11 different ratios of sucrose to fructose,their effects on sperm vitaliy were studied.The results showed that the sperm vitality of the fructose and sucrose groups was significantly better than that of the glucose and honey groups at each time point(P<0.05).After 1 hour of storage at 4℃,the sperm vitality of the fructose group was significantly higher than that of the other groups(P<0.05).After 1.5 and 2 hours of storage at 4℃,the sperm vitality in the sucrose group was significantly higher than that in the other groups(P<0.05).Additionally,the forward movement ratio of sperm in the fructose group was significantly higher than that in the other groups after 0.5 and 1 hour of storage at 4℃(P<0.05).In summary,the use of sucrose and fructose as carbon sources in the diluent achieved the best results for the cryopreservation of chicken semen.Therefore,the optimal ratio of sucrose to fructose was further explored.With the extension of storage time at 4℃,the sperm vitality of all groups showed a significant decrease(P<0.05).The forward movement ratio of sperm in group B was significantly higher than that in the other groups after 5 days of storage at 4℃(P<0.05).Except for group C,the forward movement ratio and sperm vitality in group B were significantly higher than those in the other groups after 10 days of storage at 4℃(P<0.05).Among the different ratios of fructose and sucrose,group B exhibited the best overall sperm vitality and forward movement ratio after 5 and 10 days of storage at 4℃.With the increase in the sucrose ratio,the forward movement activity of sperm decreased significantly under 4℃con
