摘 要:目的探讨松果菊苷(Ech)对口腔溃疡大鼠口腔黏膜和炎症反应的影响及其潜在的作用机制。方法30只Wistar大鼠随机分为正常对照组(NC group)、口腔溃疡模型组(Model group)、Ech组(Ech group)、口炎清颗粒治疗组(KG group)和Ech+NLRP3激活剂组(Ech+NLRP3 group),每组6只。各组大鼠进行相应的处理后,比较各组大鼠口腔溃疡面积的变化情况;ELISA检测各组大鼠炎症反应;HE染色分析各组大鼠口腔黏膜的病理变化;Western blot检测各组大鼠口腔黏膜组织中NOD样受体热蛋白结构域相关蛋白3/半胱氨酸的天冬氨酸蛋白水解酶1/白细胞介素-1β(NLRP3/Caspase-1/IL-1β)信号通路相关蛋白的表达情况。结果口腔溃疡模型大鼠的口腔溃疡面积增大,口腔黏膜病理学损伤评分升高;炎症因子IL-6、IL-1β、TNF-α水平和NLRP3、Caspase-1、IL-1β蛋白水平均上调(P<0.05)。Ech和口炎清颗粒治疗后可明显缩小口腔溃疡面积,降低口腔黏膜病理学损伤程度;下调IL-6、IL-1β、TNF-α水平和NLRP3、Caspase-1、IL-1β蛋白水平(P<0.05)。而NLRP3激活剂可部分逆转Ech治疗对口腔溃疡大鼠口腔黏膜和炎症反应的保护作用(P<0.05)。结论Ech可通过抑制NLRP3/Caspase-1/IL-1β信号通路,保护口腔溃疡大鼠免受口腔黏膜损伤和炎症反应。Objective To investigate the influences of echinacoside(Ech)on oral mucosa and inflammatory response in rats with oral ulcer and its potential mechanism.Methods Thirty Wistar rats were randomly grouped into normal control group(NC group),oral ulcer model group(Model group),Ech treatment group(Ech group),Kouyanqing granule treatment group(KG group),and Ech+NLRP3 activator group(Ech+NLRP3 group),6 rats per group.After the rats in each group were treated accordingly,the changes in the oral ulcer area of the rats in each group were compared;ELISA was applied to detect the inflammatory response of the rats in each group;HE staining was applied to analyze the pathological changes of the oral mucosa of the rats in each group;and Western blot was applied to detect the expression of NOD-like receptor thermal protein domain-associated protein 3/cysteinyl aspartate specific proteinase 1/interleukin-1β(NLRP3/Caspase-1/IL-1β)signaling pathway-related proteins in oral mucosa of rats in each group.Results In the oral ulcer model rats,the oral ulcer area increased and the oral mucosal pathological damage score increased,the levels of inflammatory factors IL-6,IL-1βand TNF-αand the protein levels of NLRP3,Caspase-1 and IL-1βwere up-regulated(P<0.05).Treatment with Ech and Kouyanqing granule significantly reduced the area of oral ulcers and the degree of pathological damage to the oral mucosa,down-regulated the levels of IL-6,IL-1βand TNF-αand the protein levels of NLRP3,Caspase-1 and IL-1β(P<0.05).NLRP3 activator could partially reverse the protective effects of Ech treatment on oral mucosa and inflammatory response in rats with oral ulcer(P<0.05).Conclusion Echinacoside can protect rats with oral ulcer from oral mucosal injury and inflammatory reaction by inhibiting the NLRP3/Caspase-1/IL-1βsignaling pathway.
