摘　　要：目的探讨钙调磷酸酶抑制剂他克莫司对柯萨奇病毒B3(CVB3)诱导的细胞自噬的作用。方法将细胞分为对照组、CVB3感染组、CVB3感染+他克莫司组,采用Western blotting分别检测LC3Ⅰ、LC3Ⅱ、p62、VP1蛋白的表达水平。采用RT-qPCR检测他克莫司对CVB3病毒基因组的吸附作用。根据半数组织感染剂量(TCID 50)判断他克莫司对CVB3病毒滴度的表达情况。结果CVB3感染诱导自噬表达水平增加,且随着他克莫司的加入,CVB3诱导的自噬呈现下降趋势。他克莫司下调CVB3病毒衣壳蛋白VP1的表达,并降低CVB3病毒基因组的吸附作用和病毒滴度。结论他克莫司可以降低CVB3病毒基因的复制,改善CVB3诱导的细胞自噬。Objective To investigate the role of tacrolimus,a calmodulin phosphatase inhibitor,on coxsackievirus B3-induced cellular autophagy.Methods The cells were divided into control group,CVB3-infected group,and CVB3-infected+tacrolimus group,and the expression levels of LC3Ⅰ,LC3Ⅱ,p62,and VP1 proteins were detected by Western blotting,respectively.RT-qPCR was used to detect the adsorption of tacrolimus on CVB3 viral genome.The expression of tacrolimus on the titer of CVB3 virus was judged according to the half tissue infectious dose(TCID 50).Results CVB3 infection induced an increase in the expression level of autophagy,and CVB3-induced autophagy showed a decreasing trend with the addition of tacrolimus.Tacrolimus down-regulated the expression of CVB3 viral capsid protein VP1 and decreased the adsorption of CVB3 viral genome and viral titer.Conclusion Tacrolimus reduces CVB3 viral gene replication and improves CVB3-induced cellular autophagy.
