摘 要:为优化木棉树皮总黄酮的提取工艺,以木棉树皮总黄酮得率为考察指标,采用减压内部沸腾法,通过响应面法优化确定木棉树皮总黄酮的最佳提取工艺。响应面优化最佳减压内部沸腾提取工艺条件为:提取压力为0.035 MPa,解吸剂乙醇体积分数为51%,提取剂乙醇体积分数41%,液料比为61∶1,提取温度为58℃,在此条件下总黄酮得率平均值达18.45%,与预测值18.59%相比,相对误差为0.75%,证明模型理论预测值与实际值拟合效果很好。以总黄酮吸附率和解吸率为考察指标,确定AB-8为最佳树脂,并得到大孔树脂纯化的最佳条件为:上样液质量浓度为0.7 mg/mL、pH值为5,温度为30℃,体积为80 mL;吸附饱和的树脂水洗后依次用20%和70%的乙醇各100 m L进行梯度洗脱。实验结果表明,优化的木棉树皮总黄酮减压提取工艺合理、可行,大孔树脂纯化后总黄酮纯度达到82.95%,是传统醇提法的3.54倍。抗氧化实验表明木棉皮总黄酮清除超氧阴离子自由基的IC50为0.31 mg/mL,说明大孔树脂纯化的木棉皮总黄酮具有较强的清除超氧阴离子自由基的能力。In order to optimize the extraction technology of total flavonoids from kapok bark,based on the total flavonoids extraction rate as the index of kapok bark,it can be drawn that the optimum extraction process of total flavonoids from kapok bark by the optimization method of combining the internal boiling method with the response surface method(RSM).The experimental results showed that the optimal extraction parameters were as follows:extraction pressure was 0.035 MPa.The desorption agent concentration was 51%.The extraction concentration was 41%.The liquid-solid ratio was 61:1 and extraction temperature was 58℃.Under these conditions,the average extraction rate of total flavonoids reached 18.45%,the relative error was 0.75%compared with the predicted value of 18.59%,which proved that the theoretical predicted value of the model fitted very well with the actual value.The adsorption rate and desorption rate of total flavonoids were used as the indexes.Resin AB-8 was determined to be the best resin through the static and dynamic adsorption experiments.While the optimal adsorption conditions showed that the concentration of sample liquid was 0.7 mg/mL and sample pH was 5 with the temperature of 30℃.The sample volume was 80 mL.The resins after adsorption were eluted with water,and then gradient elution with 20%and 70%ethanol,the volume of elution was 100 mL,respectively.The results showed that the optimization of decompressing extraction process of total flavonoids from Kapok Bark was reasonable and feasible.The purity of total flavonoids from kapok bark reached 82.95%.Compared with traditional method,the mass concentration of total flavonoids after enrichment with macroporous adsorbent resin was 3.54 times that of the traditional alcohol formulation.The IC50 of total flavonoids from Kapok Bark.after purification by AB-8 on the superoxide anions radical were 0.31 mg/mL,respectively,showing strong antioxidant activity.
