摘 要:目的考察川芎配伍天麻后对川芎嗪检出含量的影响。方法通过高效液相法,对川芎嗪单药组和不同配伍比例的药对组进行川芎嗪的含量测定。采用色谱柱Venusil MP-C_(18)(4.6 mm×250 mm,5μm);以甲醇-水(50∶50,体积比)为流动相;检测波长:280 nm;柱温25℃,流速0.8 mL·min^(-1)。结果川芎嗪含量在0.30~20μg·mL^(-1)范围内与峰面积值呈良好线性关系(r=0.9994),精密度和重复性试验的RSD均<2.0%(n=6),稳定性试验结果显示各组样品在24 h内稳定,川芎组、天麻川芎A组、天麻川芎B组川芎嗪的平均回收率分别为:101.31%、103.49%、102.26%,RSD值分别为:1.90%,1.18%,1.79%。川芎组、天麻川芎A组、天麻川芎B组的川芎嗪检出含量依次为0.0743 mg·g^(-1)、0.095 mg·g^(-1)、0.0826 mg·g^(-1)。结论天麻川芎配伍有利于川芎嗪的提取,1∶4的配伍比例优于4∶1的配伍比例。OBJECTIVE Examining the compatibility of Gastrodia elata Blume and Ligusticum chuanxiong Hort affects the quantity of ligustrezine.METHODS The content of ligustrazine was determined in the single drug group and the drug pair group with different compatibility ratios by HPLC.The chromatographic separation was performed on an chromatographic column Venusil MP-C_(18)(4.6 mm×250 mm,5μm)with methanol-water(50∶50,volume ratio)as the mobile phase.The detection wavelength was 280 nm.The column temperature was kept at 25℃ and the flow rate was 0.8 mL·min^(-1).RESULTS The content of ligustrazine showed a good linear relationship with peak area values within the range of 0.30-20μg·mL^(-1)(r=0.9994),and the RSD of precision and repeatability tests were both<2.0%(n=6).The stability test results showed that the samples in each group were stable within 24 hours.The average recovery rates of Ligusticum chuanxiong Hort group and Gastrodia elata Blume and Ligusticum chuanxiong Hort group A(4∶1)and Gastrodia elata Blume and Ligusticum chuanxiong Hort group B(1∶4)were 101.31%,103.49%,and 102.26%,respectively,with RSD values were 1.90%,1.18%,and 1.79%.The detected levels of ligustrazine content in the Ligusticum chuanxiong Hort group and Gastrodia elata Blume and Ligusticum chuanxiong Hort group A(4∶1)and Gastrodia elata Blume and Ligusticum chuanxiong Hort group B(1∶4)were 0.0743 mg·g^(-1),0.095 mg·g^(-1)and 0.0826 mg·g^(-1),respectively.CONCLUSION The Gastrodia Chuanxiong couplet medicines compatibility is conducive for ligustrazine extraction,and a 1∶4 ratio of compatibility is better than 4∶1 ratio.
