摘  要:目的探索聚乙二醇干扰素α-2b(PEG IFNα-2b)对比普通干扰素α-2b(IFNα-2b)在呼吸道合胞病毒(RSV)细胞模型中抗病毒活性及其稳定性。方法采用细胞病变(CPE)法检测不同浓度的PEG IFNα-2b和普通IFNα-2b分别在高温环境、酶切环境下的药物半抑制浓度(IC_(50))。结果对比正常环境,在高温环境下,PEG IFNα-2b的抗病毒活性维持稳定(IC_(50)维持不变),而普通IFNα-2b活性明显降低(IC_(50)增加1.9~9.6倍);酶切环境对PEG IFNα-2b的抗病毒活性略有影响(IC_(50)增加2.7~4.3倍),而对普通IFNα-2b活性有极大的影响(IC_(50)增加9.6~18.9倍)。结论PEG IFNα-2b对RSV病毒有相对稳定的抗病毒活性,为进一步的体内研究提供基础数据,有助于优化当前的抗RSV病毒策略。OBJECTIVE To investigate the antiviral activity and stability of pegylated interferon α-2b(PEG IFNα-2b)compared to conventional interferon α-2b(IFNα-2b)in a respiratory syncytial virus(RSV)cell model.METHODS Cytopathic effect(CPE)detection was used to measure the half maximum inhibitory concentration(IC_(50))of PEG IFNα-2b and conventional IFNα-2b at high temperature and under enzyme digestion.RESULTS At high temperatures,PEG IFNα-2b's antiviral activity remained steady(IC_(50)remained unaltered)as compared to a normal environment,whereas conventional IFNα-2b's activity significantly dropped(IC_(50)increased by 1.9-9.6 times).The antiviral activity of PEG IFNα-2b(IC_(50)raised by 2.7-4.3 fold)was somewhat affected by the enzyme digesting environment,whereas conventional IFNα-2b(IC_(50)increased by 9.6-18.9 fold)was greatly affected.CONCLUSION PEG IFNα-2b has relatively stable antiviral activity against RSV,providing basic data for further in vivo studies and helping to optimize current anti-RSV strategies.
