摘　　要：目的探究液基细胞样本预处理前后对HPV检测结果的影响及两种检测方法的一致性。方法选取液基细胞样本共253例,预处理前后,对比HPV阳性检出率、16/18检出率、混合型检出率及一致性情况。结果①253例样本,经预处理后HPV阳性检出率明显高于预处理前,差异有统计学意义(P<0.05),16/18检出率及混合型检出率均高于预处理前,但差异无统计学意义(P>0.05),预处理前后检测结果一致性较强,Kappa值0.739。②其中103例样本采用PCR-荧光探针法和PCR-反向点杂交法检测,预处理前后的检测结果均具有强一致性,且预处理后的一致性更高(Kappa值0.933>0.922)。结论液基细胞样本经预处理后能明显提高HPV阳性检出率,能检出更多16/18阳性数、混合型数;采用不同检测方法,预处理后的样本具有更高的一致性及稳定性。Objective To explore the influence of liquid-based cell samples before and after pretreatment on HPV detection results and the consistency of the two detection methods.Methods A total of 253 liquid-based cell samples were selected,and the positive detection rate,16/18 detection rate,mixed type detection rate and consistency of HPV were compared before and after pretreatment.Results In 253 samples,the positive detection rate of HPV after pretreatment was significantly higher than that before pretreatment,the difference was statistically significant(P<0.05),the detection rate of 16/18 and mixed type were higher than that before pretreatment,but the difference was not statistically significant(P>0.05).The detection results before and after pretreatment were consistent,and Kappa value was 0.739.103 samples were detected by PCR-fluorescent probe method and PCR-reverse dot hybridization method,and the detection results before and after pretreatment showed strong consistency,and the consistency was higher after pretreatment(Kappa value 0.933>0.922).Conclusion The pre-treatment of liquid-base cell samples can significantly improve the positive rate of HPV,and more 16/18 positive number and mixed type number can be detected.Using different detection methods,the samples after pretreatment have higher consistency and stability.
