摘 要:目的研究彝医药灵通清肺汤(Lingtong Qingfei Decoction,LTQFD)改善脂多糖(lipopolysaccharide,LPS)介导大鼠肺毛细血管渗漏(capillary leakage,CL)的潜在分子机制。方法用LTQFD干预LPS诱导的急性肺损伤(acute lung injury,ALI)大鼠,进行肺组织基因芯片分析及蛋白互作网络构建,检测肺炎性状态、CL指标和气道指标、呼吸功能和病理改变;进而用正常大鼠LTQFD含药血清预处理LPS干预的大鼠肺静脉内皮细胞(rat pulmonary vein endothelial cells,RPPVECs),检测促炎细胞因子释放、跨膜电阻和细胞旁渗漏及枢纽蛋白c-Fos的表达。结果LTQFD抑制了LPS介导的Toll样受体级联激活和c-Fos高表达,改善了过激的炎性反应,缓解了肺和RPPVECs的CL,从而改善了ALI的肺病理改变、气道指标和呼吸功能。结论LTQFD可能通过逆转LPS介导的Toll样受体级联激活和c-Fos高表达来改善ALI肺CL、病理学改变、气道指标和呼吸功能。Objective To investigate the potential molecular mechanism of Lingtong Qingfei Decoction(LTQFD)in ameliorating lipopolysaccharide(LPS)-induced pulmonary capillary leakage in rats.Methods Rats with LPS-induced acute lung injury(ALI)were treated with LTQFD.Lung tissue gene chip analysis and the construction of a protein interaction network were conducted.Subsequently,assessments were made on pneumonia status,capillary leakage,airway indicators,respiratory function,and pathological changes.LPS-intervened rat pulmonary vein endothelial cells(RPPVECs)were pre-treatment with LTQFD(normal rat serum containing medication)to evaluate pro-inflammatory cytokine release,transepithelial electrical resistance,paracellular leakage,and hub protein c-Fos expression.Results LTQFD inhibited LPS-mediated Toll-like receptor cascade activation and high expression of c-Fos,improved the excessive inflammatory response,relieved capillary leakage in the lungs and RPPVECs,and thus improved lung pathological changes,airway indicators,and respiratory function in ALI(P<0.05).Conclusion LTQFD might improve pulmonary capillary leakage,pathological changes,airway indicators,and respiratory function in ALI by reversing LPS-mediated Toll-like receptor cascade activation and high expression of c-Fos.
