摘　　要：目的分析胚胎质量与颗粒细胞线粒体DNA(mtDNA)拷贝数之间的相关性。方法选取67名2022年1月至2022年12月在医院生殖医学中心接受辅助生殖技术助孕的女性,以接受助孕女性的正常受精胚胎81个作为研究对象,根据D3卵裂期胚胎形态学评分将胚胎分为对照组(胚胎评分<3分)和观察组(胚胎评分≥3分),对女性实施控制性超促排卵后取卵,应用血清/血浆游离DNA提取试剂盒提取培养液中游离DNA并应用数字并PCR法检测mtDNA拷贝数,比较不同D3卵裂期胚胎形态学评分女性基础内分泌情况及不同胚胎评分女性妊娠结局,分析mtDNA拷贝数与胚胎评分及妊娠结局相关性。结果与对照组女性相比,观察组女性孕酮(P)、雌二醇(E_(2))、黄体生成素(LH)、卵泡刺激素(FSH)及mtDNA拷贝数、人绒毛膜促性腺激素(HCG)日内膜厚度值均更高(P<0.05)。观察组妊娠成功率较对照组高(P<0.05)。颗粒细胞线粒体mtDNA拷贝数与胚胎评分及妊娠成功率均呈正相关(P<0.05)。结论胚胎质量与颗粒细胞mtDNA拷贝数存在正相关性,检测颗粒细胞mtDNA拷贝数可为临床评估胚胎质量以及妊娠结局提供指导和参考。Objective To analyze the correlation between embryo quality and mitochondrial DNA(mtDNA)copy number in granulosa cells.Method 67 women who received assisted reproductive technology from the hospital's reproductive medicine center from January 2022 to December 2022 were selected,and 81 normal fertilized embryos from women who received assisted reproduction were selected as the research objects.According to the D3 cleavage stage embryo morphology score,the embryos were divided into a control group(embryo score<3 points)and an observation group(embryo score≥3 points).After controlled ovarian hyperstimulation,eggs were taken from the women.Serum/plasma free DNA extraction kit was used to extract free DNA from the culture medium,and mtDNA copy number was detected by digital PCR method.The basic endocrine status of women with different D3 cleavage stage embryo morphology scores and the female pregnancy outcomes with different embryo scores were compared,and the correlation between mtDNA copy number and embryo score and pregnancy outcomes was analyzed.Result Compared with the control group,the observation group had higher levels of progesterone(P),estradiol(E_(2)),luteinizing hormone(LH),follicle stimulating hormone(FSH),mtDNA copy number,and human chorionic gonadotropin(HCG)daily endometrial thickness(P<0.05).The success rate of pregnancy in the observation group was higher than that in the control group(P<0.05).The copy number of mitochondrial mtDNA in granulosa cells is positively correlated with embryo score and pregnancy success rate(P<0.05).Conclusion There is a positive correlation between embryo quality and granulosa cell mtDNA copy number.Detecting granulosa cell mtDNA copy number can provide guidance and reference for clinical evaluation of embryo quality and pregnancy outcomes.
