摘 要:超长链脂肪酸代谢合成过程中的关键限速酶KCS1对于植物抵御非生物胁迫具有重要作用,为了探究大豆KCS1基因的功能,本研究从栽培大豆中克隆出大豆KCS1基因并对该基因进行生物信息学分析以及对该基因的组织表达情况、盐胁迫下的表达变化情况进行探究。结果表明:GmKCS1基因启动子区域内与胁迫响应相关的顺式作用元件数量最多,为10个;大豆KCS1蛋白与野生大豆KCS1蛋白、拟南芥KCS1蛋白、谷子KCS1蛋白聚类为同一进化分支,大豆KCS1蛋白与野生大豆KCS1蛋白的进化关系最近;GmKCS1基因在大豆根、茎、子叶和叶组织中均可表达,但在叶组织中的表达水平最高;盐胁迫可诱导GmKCS1基因表达,GmKCS1基因相对表达水平在盐胁迫8 h和12 h时显著上升,并且在盐胁迫8 h达到峰值。本研究表明,GmKCS1基因在大豆响应盐胁迫过程中可发挥重要生物学功能,并为后续在大豆耐盐分子育种过程中应用大豆KCS基因提供参考依据。The key rate-limiting enzyme KCS1 in the metabolic synthesis process of very long chain fatty acids plays a crucial role in plants'resistance to abiotic stress.To investigate the function of soybean KCSI gene,this study cloned the soybean KCS1 gene from cultivated soybean and conducted bioinformatics analysis of the gene.The gene expression in different tissues and the expression changes under salt stress conditions were also explored.The results showed that the number of cis-elements related to stress response was the largest in the promoter region of GmKCS1 gene,which was ten.Soybean KCS1 protein,wild soybean KCS1 protein,Arabidopsis KCS1 protein,and millet KCS1 protein clustered into the same evolutionary branch,and the evolutionary relationship between soybean KCS1 protein and wild soybean KCS1 protein was the closest.GmKCS1 gene could be expressed in root,stem,cotyledon,and leaf tissues of soybean,but the expression level in leaf tissues was the highest.Salt stress could induce the expression of GmKCS1 gene.The relative expression level of GmKCS1 gene increased significantly after 8 hours and 12 hours of salt stress and reached the peak after 8 hours of salt stress.The results of this study showed that GmKCSI gene could play an important biological function in the process of soybean response to salt stress,which provided a theoretical basis for the subsequent application of soybean KCS gene in the process of soybean salt tolerance molecular breeding.
