摘 要:目的规范化制备柴胡疏肝散(Chaihu Shugan San,CSS)含药血清,并观察CSS含药血清对脂多糖(lipopolysaccharide,LPS)联合干扰素(interferon-γ,IFN-γ)诱导的小鼠单核巨噬细胞RAW264.7炎症反应的作用,为其抗炎机制的研究提供实验依据。方法SD雄性大鼠灌胃CSS制备含药血清,使用高效液相色谱-质谱联用法检测含药血清中代表性化合物浓度。体外构建LPS联合IFN-γ诱导的巨噬细胞炎症模型,分别加入CSS含药血清或空白大鼠血清,检测巨噬细胞白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-10(interleukin-10,IL-10)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、精氨酸酶1(arginase 1,ARG1)的mRNA水平,观察CSS含药血清的抗炎作用。结果CSS含药血清中α-香附酮、芍药苷等化合物含量较高。与空白大鼠血清组比较,CSS组可抑制巨噬细胞促炎因子IL-1β、IL-6、TNF-α的mRNA表达,提高抑炎细胞因子IL-10及ARG1的mRNA表达。结论CSS含药血清对LPS联合IFN-γ诱导的巨噬细胞具有一定的抗炎作用。Objective To make the standardization preparation of Chaihu Shugan San(柴胡疏肝散,CSS)-containing serum.To observe the effect of CSS-containing serum on lipopolysaccharide(LPS)combined with interferon(IFN-γ)induced inflammatory response in mouse mononuclear macrophages RAW264.7,and to provide an experimental basis for the study of its anti-inflammatory mechanism.Methods The CSS-containing serum was prepared by gavage in Sprague Dawley(SD)male rats,and the concentrations of representative compounds in the CSS-containing serum were determined by high-performance liquid chromatography-mass spectrometry(HPLC-MS).The anti-inflammatory effects of CSS were observed in an in vitro model of macrophage inflammation induced by LPS combined with IFN-γ.The mRNA levels of interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α)and arginase 1(ARG1)in macrophages were measured by adding the CSS-containing serum or normal rat serum,respectively.Results The contents of compounds such asα-coumarinone and paeoniflorin were higher in CSS-containing serum group.Compared with those of the CON group,the CSS group inhibited the mRNA expressions of macrophage pro-inflammatory factors IL-1β,IL-6 and TNF-α,and increased the mRNA expressions of anti-inflammatory cytokines IL-10 and ARG1.Conclusion Serum-containing CSS effectively reduces inflammation in LPS combined with IFN-γinduced macrophages.
