摘 要:为了提高黄花菜总黄酮得率,在乙醇体积分数、提取温度、提取时间、液料比和超声功率5个单因素实验的基础上,根据Box-Behnken实验设计对提取条件进行分析和优化,利用A-8大孔树脂对黄酮粗提物进行纯化,并对其体外抗氧化活性进行研究。结果表明乙醇分数为80%、料液比1∶15、提取时间60 min、超声功率300 W、提取温度40℃,在此条件下黄花菜总黄酮得率为11.806 mg/g。抗氧化实验结果表明,黄花菜提取物对DPPH自由基、羟基自由基、ABTS自由基、超氧阴离子自由基具有一定的清除效果和还原力,但其均低于VC。本研究为黄花菜黄酮提取工艺以及其抗氧化试验提供参考依据。In order to improve the yield of flavonoids,the extraction conditions were analyzed and optimized according to Box-Behnken experimental design based on five single factor experiments including ethanol volume fraction,extraction temperature,extraction time,liquid-solid ratio and ultrasonic power,and the antioxidant activity of flavonoids in vitro was studied.The results showed that under the conditions of ethanol fraction 80%,solid-liquid ratio 1∶15,extraction time 60 min,ultrasonic power 300 W,extraction temperature 40℃,the total flavonoids yield of day flower was 11.806 mg/mL.The results of antioxidant experiments showed that the extracts of day lily had certain scavenging effect and reducing power on DPPH free radical,hydroxyl free radical,ABTS free radical and superoxide anion free radical,but all of them were lower than VC.This study provides reference for the extraction process of flavonoids in day lily and its antioxidant experiment.
