摘 要:本文研究了通过优化的超声辅助提取(UAE)和固相萃取(SPE)方法结合液相色谱-质谱联用技术(LC-MS/MS)实现人参皂苷成分快速检测的方法。确定了最佳提取条件为:提取时间30min,温度30℃,溶剂为70%甲醇,使用C18固相萃取柱,并以甲醇作为洗脱溶剂。优化的前处理方法显著提高了人参皂苷Rb1、Re、Rg1和Compound K的提取效率和纯度。在优化的LC-MS/MS条件下,建立了各成分在10~1000ng·mL^(-1)浓度范围内的标准曲线,相关系数(R^(2))均大于0.996。方法验证结果显示,检测限(LOD)和定量限(LOQ)分别在2~3ng·mL^(-1)和6~9ng·mL^(-1)范围内,日内和日间精密度(RSD)均小于5%,回收率在96.5%~98.2%之间。人参样品成分检测结果表明,所采用的UAE和SPE前处理方法结合LC-MS/MS技术,能够实现人参皂苷的高效分离和准确定量。该检测方法为人参皂苷的质量控制和药理研究提供了技术支持。This study investigates the rapid detection of ginsenosides in ginseng through optimized ultrasonic-assisted extraction(UAE)and solid-phase extraction(SPE)methods combined with liquid chromatography-mass spectrometry(LC-MS/MS).The optimal extraction conditions were determined to be:extraction time of 30 minutes,temperature of 30℃,and 70%methanol solvent concentration,using a C18 SPE column with methanol as the elution solvent.The optimized pretreatment method significantly improved the extraction efficiency and purity of ginsenosides Rb1,Re,Rg1,and Compound K.Under optimized LC-MS/MS conditions,standard curves for each component were established in the range of 10~1000ng·mL^(-1),with correlation coefficients(R^(2))all greater than 0.996.Method validation results showed that the limits of detection(LOD)and quantification(LOQ)were in the ranges of 2~3ng·mL^(-1)and 6~9ng·mL^(-1),respectively,with intra-day and inter-day precision(RSD)less than 5%,and recovery rates between 96.5%and 98.2%.The results of ginsenoside component detection in ginseng samples indicate that the UAE and SPE pretreatment methods combined with LC-MS/MS technology can achieve efficient separation and accurate quantification of ginsenosides.The study provides technical support for the quality control and pharmacological research of ginsenosides.
