摘 要:仿岩溶碳酸氢钙(CFPK)溶液作为一种新型的环境友好型改性材料,在改性分散土、膨胀土以及固化风积沙等方面已有显著成果,但制备时间较长限制了其制备速率和改性效率。基于碳酸酐酶的催化机理,选用碳酸酐酶作为催化剂,进一步提升CFPK溶液的改性效率。通过细菌培育试验、酶活测定试验、电导率试验以及Ca^(2+)浓度测定试验等,探究含碳酸酐酶菌生长和产酶活性的最优培育条件及其对CFPK溶液制备速率的影响,提出一种新型土体改性材料的制备方法。试验表明,培养温度30℃、培养基初始pH值7.2、菌液接种量6%(V/V)、恒温震荡培养箱转速120 r/min为产碳酸酐酶菌培育生长和产酶活性的最优条件,培育传代时间为32 h。此外,碳酸酐酶可以将CFPK溶液的制备速率提升8~10倍,制备时选择反应釜内二氧化碳压强为0.8 MPa,菌水体积比为0.4%。As a new environmentally friendly modified material,the Calcium Bicarbonate From Pesudo-karstification(CFPK)solution has achieved significant results in modifying dispersed soil,expansive soil,and solidified aeolian sand.However,the long preparation time limits its preparation rate and modification efficiency.Based on the catalytic mechanism of carbonic anhydrase,carbonic anhydrase was selected as the catalyst to further enhance the modification efficiency of CFPK solution.The optimal cultivation conditions for the growth and enzyme activity of carbonic anhydrase containing bacteria and their effects on the preparation rate of CFPK solution were explored through bacterial growth tests,enzyme activity assays,conductivity assays,and Ca^(2+)concentration assays.A new method for preparing soil modified materials was proposed.The results are as follows.The optimal conditions for the growth and enzyme activity of carbonic anhydrase producing bacteria were cultivation temperature of 30℃,initial pH value of the culture medium of 7.2,inoculation volume of 6%(V/V),and constant temperature shaking incubator speed of 120 r/min.The cultivation passage time was 32 hours.In addition,Carbonic anhydrase can increase the preparation rate of CFPK solution by 8~10 times.When preparing,choose a carbon dioxide pressure of 0.8 MPa and a bacterial water volume ratio of 0.4% in the reaction vessel.
