摘 要:植食性昆虫肠道细菌的组成与其发育阶段密切相关,并且能为宿主昆虫提供多方面的功能。然而番茄潜叶蛾Tuta absoluta(Meyrick)肠道细菌与发育龄期相关研究鲜有报道,本研究旨在探究番茄潜叶蛾不同龄期幼虫肠道细菌多样性特征及预测其功能。本研究选取番茄潜叶蛾1~4龄幼虫,提取幼虫肠道细菌基因组DNA,利用Illumina MiSeq二代高通量测序技术,分析其肠道细菌多样性和丰度。Alpha多样性分析显示,番茄潜叶蛾1龄幼虫肠道细菌多样性和丰富度最高,且肠道细菌丰富度随龄期增加而降低。番茄潜叶蛾4个龄期肠道细菌的优势门均为变形菌门Proteobacteria,除了4龄幼虫的优势菌属是肠杆菌属Enterobacter以外,其他龄期幼虫的优势菌属是沃尔巴克氏体属Wolbachia。PICRUSt2基因功能预测显示番茄潜叶蛾幼虫肠道细菌参与碳水化合物、氨基酸、能量的代谢,表明其可能在食物消化吸收和营养供给等方面发挥重要作用,其中不同龄期肠道细菌新陈代谢Metabolism功能有显著差异,1龄幼虫功能基因比例明显高于3龄、4龄。研究结果可为探索番茄潜叶蛾肠道细菌与宿主的相互作用奠定基础,也为番茄潜叶蛾的适应性进化和综合治理提供更多的视角和方向。The gut bacteria composition in phytophagous insects is closely linked to their developmental stages and can fulfill multiple functions for the host insect.However,few studies have investigated the relationship between gut bacteria and developmental stages in the tomato leaf miner moth,Tuta absoluta(Meyrick).This study aimed to characterize the diversity of gut bacteria in larvae of different instar stages of T.absoluta and to predict the functional roles of these bacteria.We extracted genomic DNA from the gut bacteria of T.absoluta larvae at the 1st,2nd,3rd,and 4th instar stages,and analyzed the diversity and abundance of the gut bacteria using Illumina MiSeq II high-throughput sequencing technology.Alpha diversity analysis indicated that gut bacterial diversity and abundance peaked in the 1st instar larvae of T.absoluta and diminished as instar age increased.The dominant phylum of gut bacteria across all four instar stages was Proteobacteria.Excepting that Enterobacteria was the dominant genus in the 4th instar larvae,whereas Wolbachia was predominant in the other instars.Predicted functional analysis based on the PICRUSt2 gene database revealed that the gut bacteria in T.absoluta larvae participated in carbohydrate,amino acid,and energy metabolism,suggesting their crucial roles in digestion,absorption,and nutrient provision.Among them,there were significant differences in the metabolism fuction of intestinal bacteria at different instars and the 1st instar larvae were significantly higher than that in the 3rd and 4th instars.This study provides a foundational basis for exploring the interactions between gut bacteria and their host in T.absoluta and offers additional insights and directions towards the adaptive evolution and integrated management of this pest.
