孕期砷暴露在大鼠胎盘屏障的转运及对胎盘的影响

摘  要:目的探讨胎盘对砷的屏障作用,以及砷对胎盘的影响。方法选取7周龄大鼠随机分为对照组、低、中、高剂量组,雌鼠受孕后实验组开始给予砷暴露。持续暴露20 d后,取母体血、胎鼠血通过双通道原子荧光光谱仪检测血中的砷浓度,采用HE染色做病理切片观察胎盘的病理改变,采用免疫组化观察胎盘中的胰岛素样生长因子1(Insulin-Like Growth Factor,IGF1)和胎盘生长因子(Placental Growth Factor,PLGF)阳性表达情况。结果砷暴露期间发生大鼠死亡及流产现象;各组大鼠染毒后体重随怀孕天数逐渐增加,但差异无统计学意义(P>0.05);母体与胎鼠血中砷浓度随着各剂量组增加而增加,各剂量组的胎盘对砷的透过率随着染砷浓度的增加而升高(P<0.05);与对照组相比,高剂量组胎盘质量有所下降(P<0.05),胎鼠体重随着染砷剂量增加而降低,高剂量组体重下降(P<0.05);与对照组相比,随着染砷浓度的增加胎盘海绵滋养层模糊,血窦及母体血管减少。与对照组相比,低剂量组IGF1阳性表达增高(P<0.05),中、高剂量组PLGF阳性表达增高(P<0.05)。结论随着砷处理浓度的增加,母体血中砷浓度逐渐增加,胎盘对砷的屏障作用逐渐减弱,进入到胎儿血中,导致胎盘和胎儿质量下降,胎盘中PLGF表达水平出现异常,胎盘发生病理性改变。Objective To explore the role of the placenta as a barrier to arsenic and the effects of arsenic on the placenta.Methods 7 weeks old rats were selected and randomly divided into control,low,medium and high dose groups,and the experimental group was given arsenic exposure after the female rats were conceived.After continuous exposure for 20 days,maternal and fetal rat blood were taken to detect the arsenic concentration in blood by dual-channel atomic fluorescence spectrometry,HE staining was used to make pathological sections to observe the pathological changes of the placenta,and immunohistochemistry was used to observe the positive expression of insulin-like growth factor 1(IGF1)and placenta growth factor(PLGF)in the placenta.Translated with DeepL.com(free version)placenta.Results Rat deaths and abortions occurred during arsenic exposure.The body weight of rats in each group gradually increased with the number of days of pregnancy after exposure,but there were no statistically significant differences(P>0.05).Maternal and fetal rat blood arsenic concentrations increased with the increase of each dose group,and the transmission rate of arsenic to the placenta increased with the increase of dyed arsenic concentration in each dose group,and there were statistically significant differences(P<0.05).Compared with the control group,the quality of placenta decreased in the high-dose group,and there were statistically significant differences(P<0.05).The body weight of fetal rats decreased with the increase of arsenic dyeing dose,and the body weight decreased in the high dose group,and there were statistically significant differences(P<0.05).The placental spongy trophoblastic layer was blurred and blood sinusoids and maternal blood vessels were reduced with increasing arsenic-stained concentration compared to the control group.Compared with the control group,the positive expression of IGF1 was increased in the low-dose group(P<0.05),and the positive expression of PLGF was increased in the medium-and high-dose groups(P<0.0
