摘 要:结合静电作用原理,建立了一种免电极修饰的电化学适配体传感法用于大田软海绵酸(okadaicacid,OA)的检测。首先将核酸适配体与标记信号分子的DNA杂交成双链探针,OA能与适配体特异性结合,诱发DNA探针解离释放出含信号分子的短序列DNA,导致探针分子与带电电极之间静电排斥力减弱,引起电化学信号增强,从而建立检测OA的新方法。在最优条件下,传感器在0.05~5.0 ng/mL的OA浓度之间呈现良好的线性范围,检出限为0.042 ng/mL。将该方法应用于贝类样品的加标检测,获得较好结果。An immobilization-free electrochemical aptasensor was established for the detection of okadaic acid(OA)based on electrostatic repulsion.First,the aptamer was hybridized with DNA labeled with electroactive molecules to form the probe.OA can specifically bind to the aptamer.inducing the dissociation of DNA probes to release short DNA signal molecules,leading to a weakening of the electrostatic repulsion between the probe and the charged electrode,causing an enhancement of electrochemical signals,thus establishing a new method for detecting OA.Under optimal conditions,the aptasensor showed a good linear range between 0.05~5.0 ng/mL concentration of OA,with a detection limit of 0.042 ng/mL.The method was applied to spiked testing of shellfish samples and obtained good results.
