摘  要:目的比较两种中和试验检测血清新冠病毒中和抗体的相关性和一致性,为建立假病毒中和抗体检测方法提供数据支持。方法采用微量中和试验和基于Huh-7细胞假病毒中和试验两种方法,检测本实验室保存的20份新冠病毒核酸阳性和10份新冠病毒核酸阴性的人血清的中和抗体,用SPSS 22.0和GraphPad Prism 10.1.2软件对检测结果进行相关性、一致性和特异性分析。结果微量中和试验和假病毒中和试验的相关系数r_(s)=0.886,P<0.05;微量中和试验和假病毒中和试验的Kappa系数为0.634,P<0.05。假病毒中和试验中使用的新冠病毒假病毒能特异性识别中和抗体,灵敏度为92.86%。结论微量中和试验和假病毒中和试验具有良好的相关性,两种方法的检验结果一致性较好。Objective To compare the correlation and consistency of two neutralization tests for detecting SARS-CoV-2 neutral-izing antibody,and to provide data support for establishing a method for detecting pseudovirus neutralizing antibody. Methods The neutralizing antibody of 20 SARS-CoV-2 nucleic acid positive and 10 SARS-CoV-2 negative human sera were detected by microneutralization test and Huh-7 cell pseudovirus neutralization test. SPSS 22.0 and GraphPad Prism 10.1.2 softwares were used to analyze the correlation,consistency and specificity of the detection results. Results The correlation coefficient between microneutralization test and pseudovirus neutralization test was 0.886(P<0.05). Kappa value between microneutralization test and pseudovirus neutralization test was 0.634(P<0.05). SARS-CoV-2 pseudovirus used in the pseudovirus neutralization test could specifically recognize neutralizing antibody with a sensitivity of 92.86%. Conclusion There was a good correlation between microneutralization test and pseudovirus neutralization test,the results of the two methods were agreeable.
