摘  要:锌是水生动物所必需的元素之一,且与鱼体脂肪代谢密切相关。本研究以初始体质量(5.26±0.12)g的大口黑鲈(Micropterus salmoides)为实验对象,用锌水平分别为50.1 mg/kg(Zn50)、73.3 mg/kg(Zn73)和209.2 mg/kg(Zn209)的3种饲料进行为期10周的养殖,研究不同锌水平对大口黑鲈肠道脂肪沉积以及脂代谢相关酶和基因表达的影响,以期为大口黑鲈肠道健康调节提供理论基础。实验结果表明:Zn50组、Zn73组和Zn209组的肠道脂肪含量分别为6.43%、2.74%、10.62%,各组间差异显著(P<0.05);肠道脂肪合成相关的FAS酶活性和ACC酶活性以及与脂肪分解相关的HSL酶活性均随饲料中锌含量的增加而逐渐上升,Zn50组最低,Zn73组其次,Zn209组最高(P<0.05),CPT1的活性则是在Zn73组显著高于Zn50组和Zn209组(P<0.05);相比Zn50组,Zn209组显著上调了相关基因的表达(P<0.05),包括脂肪合成相关基因(acca、fasn、srebp-1、6pgd)、脂肪分解相关基因(cpt1ab、hsla、hslb)和肠型脂肪酸结合蛋白基因(ifabp-2),而Zn73组下调了fasn基因表达,并显著上调了另外7个基因表达水平(P<0.05);相比Zn73组,Zn209组fasnf、abp-2、acca、6pgd、srebp-1、hsla基因的表达水平上调,而hslb和cpt1ab基因的表达水平显著下调(P<0.05)。综上所述,本研究中大口黑鲈饲料中的适宜锌水平为73.3 mg/kg,可通过调控大口黑鲈脂代谢相关酶活性和基因的表达,有效避免肠道中的脂肪过度沉积。Zinc is one of the essential elements in aquatic animals,which is also closely related to fat metabolism.Therefore,the aim of this study was to investigate the effect of 3 dietary zinc level,i.e.50.1 mg/kg(Zn50),73.3 mg/kg(Zn73),and 209.2 mg/kg(Zn209),on the intestinal lipid content,the enzymes activities and genes expression related to intestinal lipid metabolism of largemouth bass Micropterus salmoides with an initial weight of(5.26±0.12)g following a 10-week feeding trial.The results showed that the intestinal lipid content in the Zn50 group,Zn73 group,and Zn209 group was 6.43%,2.74%,and 10.62%,respectively,which was significantly different from each other(P<0.05).The activities of intestinal FAS,ACC and HSL increased significantly with dietary Zinc level,while CPT1 in Zn73 group was significantly higher than that in the Zn50 group and Zn209 group(P<0.05).Compared with the Zn50 group,Zn209 group significantly increased the expression of all tested genes in the intestine(P<0.05),including genes involved in lipogenesis(acca,fasn,srebp-1,and 6pgd),lipolysis(cpt1ab,hsla,and hslb)and intestinal fatty acid binding protein gene (ifabp-2), while Zn73 group down-regulated the expression of fasn gene, and significantly up-regulated the expression of other 7 genes (P < 0.05). Compared with Zn73 group, the Zn209 group significantly up-regulated the expression of ifabp-2, acca, 6pgd, srebp-1, and hsla, but down-regulated the expression of hslb and cpt1ab (P < 0.05). In conclusion, the optimal dietary zinc level of Micropterus salmoides is 73.3 mg/kg in this study to avoid excess of intestinal lipid deposition by regulating lipid metabolism.
