北方灵芝属栽培种质遗传多样性评价研究

摘  要:以来自黑龙江、吉林、辽宁、北京、山东等地39株灵芝属菌株为试材,采用ITS序列测定、体细胞不亲和性试验和SRAP分子标记方法进行遗传多样性分析,确定其种间划分及系统发育关系,以期为我国北方地区灵芝属栽培种质的分类、亲缘关系和遗传背景提供参考依据。结果表明:供试的39个灵芝属菌株包括赤芝Ganoderma lingzhi、紫芝G.sinense、松杉灵芝G.tsugae、无柄灵芝G.resinaceum和有柄灵芝G.gibbosum 5个种,相互之间有拮抗反应共713对,无拮抗反应28对,遗传相似系数(GS)为0.50~0.93,平均值为0.69。综上可知,我国北方灵芝属栽培种质存在种类丰富,遗传背景差异大,遗传多样性强的特征,但部分栽培菌株之间无拮抗反应且遗传距离很近,存在着同物异名现象。Taking 39 strains of Ganoderma species from Heilongjiang,Jilin,Liaoning,Beijing,Shandong and other places as experimental materials,the genetic diversity was analyzed by ITS sequence determination,somatic cell incompatibility test and SRAP molecular marker method to determine the species classification and systematic relationship,in order to provide reference for the classification,genetic relationship and genetic background of Ganoderma cultivar resources in northern China.The results showed that the 39 strains of Ganoderma genus tested included 5 species,Ganoderma lingzhi,G.sinense,G.tsugae,G.resinacium and G.gibbosum,and there were 713 pairs of antagonistic reactions and 28 pairs of non-antagonistic reactions among 39 strains,and the genetic similarity coefficient (GS) of the tested strains ranged from 0.50 to 0.93,the average value was 0.69.In conclusion, Ganoderma genus germplasm resources in northern China had significant differences in genetic background and strong genetic diversity,but there were also synonymous phenomena.
