摘 要:以灯盏幼苗为试材,将其分为对照(未经低温驯化)和冷驯处理(在5℃下驯化72 h获得低温驯化),2组分别接受冷冻处理(-5℃处理3 h)、融化处理(4℃处理24 h)、恢复1 d(室温处理24 h)以及恢复2 d(室温处理48 h),研究了低温对灯盏花耐寒性的积极作用,以期为提高灯盏花的抗寒性提供参考依据。结果表明:1)经过冷驯化处理后,灯盏花叶绿素b和总叶绿素含量降低;2)相对电导率显著增加38.57%,但处理在冷冻阶段、融化阶段、恢复1 d阶段和恢复2 d阶段相对电导率分别降低52.33%、71.31%、43.90%和28.40%;3)经过低温驯化后,灯盏花幼苗的可溶性蛋白质含量增加38.53%,而可溶性糖含量下降37.14%,脯氨酸含量无显著变化;4)谷胱甘肽还原酶、过氧化氢酶和抗坏血酸过氧化物酶活性在冷驯化后分别减少56.89%、91.02%和78.11%。综上所述,低温驯化能够提升灯盏花的生理调节能力,减轻其细胞膜的受损程度,从而增强其对低温胁迫的抵抗能力。Taking Erigeron breviscapus seedlings as experimental materials,they were divided into control group(without low-temperature acclimation)and cold acclimation group(acclimated at 5℃ for 72 hours).The two groups were treated with freezing treatment(-5℃ for 3 hours),melting treatment(4℃ for 24 hours),recovery for 1 day(room temperature for 24 hours)and recovery for 2 days(room temperature for 48 hours),respectively.The positive effects of low temperature on cold resistance of Erigeron breviscapus were studied,in order to provide reference for improving the cold resistance of Erigeron breviscapus.The results showed that,1)the chlorophyll b and total chlorophyll contents of Erigeron breviscapus were reduced after cold domestication;2)the relative conductivity was significantly increased by 38.57%,but the relative conductivities of the treatment groups were reduced by 52.33%,71.31%,43.90%,and 28.40%in the freezing,thawing,recovery 1 day and recovery 2 days stages,respectively;3)after low-temperature domestication,the soluble protein content of Erigeron breviscapus seedling increased by 38.53%,while soluble sugar content decreased by 37.14%,and there was no significant change in proline content;4)glutathione reductase,catalase,and ascorbate peroxidase activities decreased by 56.89%,91.02%,and 78.11%,respectively,after cold domestication.In conclusion,low-temperature domestication was able to enhance the physiological regulation ability of Erigeron breviscapus,reduce the degree of damage to its cell membrane,and thus enhance its resistance to low-temperature stress.
