摘 要:目的探讨cpsG基因对神经胶质瘤细胞U87MG转录组学影响。方法将慢病毒rLV-cpsG感染人胶质瘤细胞U87MG,筛选稳定细胞系。提取细胞总RNA,进行高通量RNA测序分析。包括差异基因分析、GO分析、KEGG分析、Reactome分析、DO分析、差异基因的可变剪切分析等。结果通过高通量RNA测序技术发现cpsG影响了U87MG细胞的基因转录能力,其中上调的基因有TMEM132A、CRAMP1、PRSS3、MAP4K3、SEMA3B等;下调的基因有HOXA11、NFYA、ICA1、SLC7A2、SOX8等。KEGG分析发现上调的KEGG通路主要包括:PI3K-Akt信号通路、Toll样受体信号通路、TNF信号通路、NF-kappa B信号通路;下调的KEGG通路主要包括:Wnt信号通路、MAPK信号通路、cAMP信号通路、Hippo信号通路。Reactome分析发现上调的Reactome通路主要包括:细胞外基质组织、蛋白质代谢、止血、免疫系统;下调的Reactome通路主要包括:神经系统、生物发育、信号转导、肌肉收缩。DO分析发现上调的DO通路主要包括:鳞状细胞癌、胸部癌症、生殖器官癌、腺癌、胃癌;下调的DO通路主要包括:中枢神经系统癌症、精神病性障碍、认知障碍、卵巢透明细胞腺癌。可变剪切分析发现:SE占65.97%,MXE占8.07%,A5SS占8.68%,RI占6.82%,A3SS占10.46%。结论cpsG基因调控了神经胶质瘤细胞的转录组学,为人神经胶质瘤的临床诊断和治疗提供了理论依据。Objective To investigate the effect of cpsG gene on the transcriptome of glioma cell line U87MG.Methods Lentivirus rLV-cpsG was infected with human glioma cell U87MG,and the stable cell line was screened.Total RNA was extracted and analyzed by high-throughput RNA sequencing.It included differential gene analysis,GO analysis,KEGG analysis,Reactome analysis,DO analysis,variable shear analysis of differential genes,etc.Results High-throughput RNA sequencing technology showed that cpsG affected gene transcription ability of U87MG cells,and upregulated genes included TMEM132A,CRAMP1,PRSS3,MAP4K3,SEMA3B,etc;down-regulated genes included HOXA11,NFYA,ICA1,SLC7A2,SOX8 and so on.KEGG analysis showed that upregulated KEGG pathways mainly included PI3K-Akt signaling pathway,Toll-like receptor signaling pathway,TNF signaling pathway,NF-kappa B signaling pathway,etc;the down-regulated KEGG pathways mainly included Wnt signaling pathway,MAPK signaling pathway,cAMP signaling pathway,Hippo signaling pathway,etc.Reactome analysis showed that the up-regulated Reactome pathways mainly included extracellular matrix organization,protein metabolism,hemostasis,immune system,etc;the down-regulated Reactome pathways mainly included nervous system,biological development,signal transduction,muscle contraction,etc.DO analysis showed that the up-regulated DO pathway mainly included squamous cell carcinoma,chest cancer,genital organ cancer,adenocarcinoma,gastric cancer,etc;the down-regulated DO pathway mainly includes central nervous system cancer,psychiatric disorders,cognitive disorders,and ovarian clear cell adenocarcinoma.Variable shear analysis showed that SE accounted for 65.97%,MXE accounted for 8.07%,A5SS accounted for 8.68%,RI accounted for 6.82%,and A3SS accounted for 10.46%.Conclusion cpsG gene regulates the transcriptomics of glioma cells,which provides a theoretical basis for the clinical diagnosis and treatment of human glioma.
