金华市汉族献血人群Lutheran、Duffy、Kidd、Diego、Dombrock5个血型系统基因频率及多态性分析

摘　　要：目的对金华市汉族献血人群Lutheran、Duffy、Kidd、Diego、Dombrock 5个稀有血型系统进行基因分型,分析主要抗原的基因分布频率。方法回顾性采用实时荧光定量聚合酶链反应对金华市中心血站2023年2至10月无偿献血者91例汉族患者血液标本的5个稀有血型系统进行基因分型和多态性分析,比较金华市和其他地区5个稀有血型系统的基因频率和分布的差异。结果91例无偿献血者Lutheran、Duffy、Kidd、Diego、Dombrock 5个型系统等位基因频率分别是Au^(a)=0.918、Au^(b)=0.082;Fy^(a)=0.967、Fy^(b)=0.033;Jk^(a)=0.407、Jk^(b)=0.593;Di^(a)=0.017、Di^(b)=0.984;Do^(a)=0.066、Do^(b)=0.934。5个血型系统对偶抗原的不配合率分别为13.98%、6.18%、36.61%、3.19%、11.55%且均符合Hardy-Weinberg平衡(均P>0.05)。金华市汉族献血人群Au^(a)、Au^(b)等位基因频率分布与海南黎族自治县黎族、新疆维吾尔自治区维吾尔族人群比较,差异均有统计学意义(均P<0.05);金华市汉族献血人群Fy^(a)、Fy^(b)等位基因频率分布与新疆维吾尔自治区回族、新疆维吾尔自治区维吾尔族、新疆维吾尔自治区哈萨克族、运城市汉族、西安省汉族人群比较,差异均有统计学意义(均P<0.05);金华市汉族献血人群Jk^(a)、Jk^(b)等位基因频率分布与新疆维吾尔自治区维吾尔族、新疆维吾尔自治区哈萨克族、西藏自治区藏族、四川省彝族、哈尔滨市满族人群比较,差异均有统计学意义(均P<0.05);金华市汉族献血人群Di^(a)、Di^(b)等位基因频率分布与岳阳市汉族人群比较,差异均有统计学意义(均P<0.05);金华市汉族献血人群Do^(a)、Do^(b)基因频率分布与新疆维吾尔自治区维吾尔族、新疆维吾尔自治区哈萨克族、西藏自治区藏族人群比较,差异均有统计学意义(均P<0.05)。结论金华市汉族献血人群稀有血型系统基因频率分布呈现遗传多态性,等位基因对偶抗原不配合率较高,与我国Objective To conduct genotyping of five rare blood group systems,namely Lutheran,Duffy,Kidd,Diego and Dombrock in Han blood donors in Jinhua area,and analyze the gene distribution frequencies of the main antigens.Methods Real-time fluorescence quantitative polymerase chain reaction was used to conduct genotyping and polymorphism analysis on the five rare blood group systems from blood samples of 91 Han voluntary blood donors at Jinhua Blood Central from February to October 2023.The differences in gene frequencies and distributions of the five rare blood group systems between Jinhua and other regions were compared.Results The allele frequencies of the five blood types Lutheran,Duffy,Kidd,Diego,and Dombrock in 91 voluntary blood donors were:Au^(a)=0.918,Au^(b)=0.082;Fy^(a)=0.967,Fy^(b)=0.033;Jk^(a)=0.407,Jk^(b)=0.593;Di^(a)=0.017,Di^(b)=0.984;Do^(a)=0.066,Do^(b)=0.934.The non-matching rates of paired antigens in the five blood type systems were 13.98%,6.18%,36.61%,3.19%,and 11.55%,respectively,and all followed the Hardy-Weinberg genetic law(all P>0.05).The frequency distribution of Aua and Aub alleles among Han population in Jinhua area showed statistically significant differences compared to the Li ethnic group in Li Autonomous Hainan Province and the Uyghur ethnic group in Xinjiang Uyghur Autonomous Region(both P<0.05);the frequency distribution of Fy^(a) and Fy^(b) alleles in Han population in Jinhua area showed statistically significant differences compared to the Hui,Uyghur,and Kazak ethnic groups in Xinjiang Uyghur Autonomous Region,and Han in Yuncheng and Xi'an(all P<0.05);compared with the Uyghur and Kazak ethnic groups in Xinjiang Uyghur Autonomous Region,the Tibetan ethnic group in the Tibet Autonomous Region,the Yi ethnie group in Sichuan and the Manchu ethnie group in Harbin,the frequency distribution of Jk^(a) and Jk^(b) alleles in Han population in Jinhua area was statistically significant(all P<0.05);the frequency distribution of Di^(a) and Di^(b) alleles in Jinhua Han population was significantly di
