两种石灰岩树种丛枝菌根真菌多样性的比较研究

摘  要:通过比较分析青檀和银缕梅根部丛枝菌根真菌的群落组成和多样性,初步探究青檀和银缕梅两种石灰岩树种的根内丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)群落组成与根际土壤丛枝菌根真菌群落组成联系和分布特征差异。以生境相似的第三纪孑遗植物青檀和银缕梅的根和根际土壤为研究对象,采用Illumina MiSeqTM高通量测序技术对两个树种的根内共生AMF种类和根际土壤AMF群落进行测序并进行相关分析。结果表明:两种石灰岩树种的根内和根际土壤的AMF都以球囊菌门为优势菌群。纲水平上青檀根内和根际土壤都以球囊菌纲为第一优势菌群,而类球囊菌纲在银缕梅根内和根际土壤中占绝对优势。目水平上球囊霉目在青檀根内和根际土中分别约占47.84%、42.51%,类球囊霉目在银缕梅根内和根际土中分别占比60.28%、59.45%。科水平上青檀根内和根际土壤的AMF未分类其他菌科占比最多,根内AMF球囊霉科约占29.98%,其次是类球囊霉科占比28.21%,近明球囊霉科占5.37%。而银缕梅根内类球囊霉科约占60.28%,其次是其他未分类菌群和少量球囊霉科菌群。青檀根际土壤中类球囊霉科约占33.78%,球囊霉科占比19.24%。相比青檀根际土壤,银缕梅根际土壤中类球囊霉科占比较多,球囊霉科占比则较少。属水平上青檀根内AMF球囊霉属占比多于类球囊霉属,根际土中则相反。银缕梅根内和根际土壤AMF则都以类球囊霉属为优势属。通过AMF群落多样性分析,两个树种根际土壤AMF丰富度和多样性高于根内,种内组成相似且种间AMF群落组成具有差异性。初步探究得出青檀和银缕梅两石灰岩山地树种根际中均存在AMF菌群且都以球囊菌门为优势菌群,但种间AMF群落组成存在差异性,种内根内和根际土的AMF群落组成相似且存在差异,可能与其分布格局和树种密切相关。By making a comparative analysis of the community composition and diversity of AMF in the roots of Pteroceltis tatarinowii and Parrotia subaequalis,the relationship and distribution characteristics difference between the community composition of AMF in the roots and thizosphere soil of P.tatarinowii and P.subaequalis were studied.The study took the roots and rhizosphere soil of Tertiary relic plants P.tatarinowii and P.subaequalis which have the similar habitats as its research object and employed Illumina MiSeg TM high-throughput sequencing technology to sequence and analyze the symbiotic AMF types in the roots and AMF communities in the rhizosphere soil of the two tree species.The results showed that Glomeromycota was the dominant flora in AMF in the roots and rhizosphere soil of the two limestone tree species.At the phylum level,Glomeromycota was the dominant flora of both limestone species.At the class level,Glomeromycetes were the first dominant fungal community in both the root and rhizosphere-soil of P.tatarinowii,while Paraglomeromycetes were the absolute predominance in P.subaequalis.At the order level,Glomerales accounted for 47.84%and 42.51%in the root and rhizosphere-soil of P.tatarinowii,while Paraglomerales accounted for 60.28%and 59.45%in the root and rhizospheresoil of P.subaequalis.At the family level,the AMF in the root and rhizosphere-soil of P.tatarinowii was the most in other fungal community.AMF Glomeraceae accounted for 29.98%of the root followed by Paraglomeraceae(28.21%)and Claroideoglomeraceae(5.37%).About 60.28%of Paraglomeraceae were in the root of P.subaequalis,followed by other unclassified fungal community and a small number of Glomeraceae fungal community.In the rhizosphere soil of P.tatarinowii,Paraglomeraceae accounted for 33.78%and Glomeraceae accounted for 19.24%.Compared with the rhizosphere soil of P.tatarinowii,the rhizosphere soil of P.subaequalis had more Paraglomeraceae and less Glomeraceae.At the genus level,the proportion of Glomus in the root was higher than Paraglomus
