我国特有濒危药用植物华山参叶片组培成苗技术研究

摘  要:目的:通过华山参(Physochlaina infundibularis)叶片培养获得再生植株,以期实现华山参植物组织培养种苗繁育,为华山参人工栽培提供种苗。方法:以华山参叶片为外植体,选用双子叶植物常用植物生长调节剂6-BA和NAA,探究脱分化、芽分化、芽生长和根分化各阶段的适宜培养基,再通过炼苗移栽得到再生植株。结果:华山参叶片脱分化诱导愈伤组织的适宜培养基为:MS+1.0 mg/L 6-BA+0.01 mg/L NAA;不定芽分化诱导的适宜培养基为:MS+8.0 mg/L 6-BA+0.01~0.08 mg/L NAA;不定芽生长培养的适宜培养基为:MS+4.0 mg/L 6-BA+0.01 mg/L NAA;经过生长培养的不定芽在不添加植物激素的1/2 MS培养基上培养60 d生根得到无菌苗;无菌苗在饥饿培养条件下,芽、叶自然凋亡,营养物质转移积累于根部形成肉质根,经过程序性炼苗移栽即可获得再生植株。结论:华山参叶片可作为适宜的繁殖材料,通过植物组织培养能够获得生长良好的再生植株。Objective:To obtain regenerated plants through leaf tissue culture of Physochlaina infundibularis,in order to establish a tissure culture propagation systerm for mass production of highquality seedings to support its artificial cultivation.Methods:The leaves were used as explants,with 6-BA and NAA,the common plant growth regulators of dicotyledonous plants,were selected to investigate the suitable medium for dedifferentiation,bud differentiation,bud growth and root differentiation.Subsequently,the regenerated plants were obtained by hardening seedlings and transplanting.Results:The suitable medium for dedifferentiation of P.infundibularis leaves to form callus was MS+1.0 mg/L 6-BA+0.01 mg/L NAA.The suitable medium for inducing adventitious bud was MS+8.0 mg/L 6-BA+0.01-0.08 mg/L NAA.The suitable medium for adventitious bud growth was MS+4.0 mg/L 6-BA+0.01 mg/L NAA.Adventitious buds were cultured on 1/2 MS medium without plant hormones for 60 days,and the sterile seedlings were obtained.Under the condition of starvation culture,the buds and leaves of the sterile seedlings withered naturally,and nutrients were transferred and accumulated in the roots,forming fleshy roots.Regenerated plants could be obtained by programmed acclimatization and transplantation of seedlings.Conclusions:P.infundibularis leaves could be used as suitable propagating materials,and the regenerated plants with good growth could be obtained by plant tissue culture.
