基于网络药理学的栀子厚朴汤治疗抑郁、焦虑和失眠的作用机制研究

摘  要:目的:采用网络药理学探究栀子厚朴汤治疗抑郁、焦虑和失眠的作用靶点及信号通路,进一步探讨其治疗抑郁、焦虑和失眠的作用机制。方法:采用中药系统药理学数据库与分析平台(TCMSP)及文献挖掘整理栀子厚朴汤化学成分,从PharmMapper获取活性成分的靶点,利用GeneCards、OMIM、Drugbank、TTD数据库收集抑郁、焦虑和失眠的相关靶点,利用Venn工具筛选交集靶点,利用STRING数据库和Cytoscape 3.7.2软件构建交集靶点蛋白质-蛋白质相互作用(PPI)网络,筛选核心靶点,通过Metascape进行基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析,利用Cytoscape 3.7.2软件构建成分-靶点-通路网络。结果:筛选出栀子厚朴汤43个有效活性成分,对应靶点290个;抑郁、焦虑和失眠疾病靶点分别有1 323个、2 765个、412个;经Venn数据库预测到疾病-药物交集靶点分别有137个、165个、68个;核心活性成分有槲皮素、山柰酚、木犀草素、β-谷甾醇、厚朴新酚等;KEGG富集显示白细胞介素(Interleukin,IL)-17、肿瘤坏死因子(Tumor Necrosis Factor,TNF)、糖尿病并发症中的晚期糖基化终产物及其受体(Advanced Glycation End Products-Receptor Advanced Glycation End Products,AGE-RAGE)等为栀子厚朴汤治疗抑郁、焦虑和失眠的主要通路。结论:该研究初步揭示栀子厚朴汤可通过多成分、多靶点、多通路治疗抑郁、焦虑和失眠,为栀子厚朴汤的深入临床研究提供了理论基础。Objective: To explore the target and signal pathway of Zhizi Houpo decoction( 栀 子 厚 朴 汤) in the treatment of depression, anxiety and insomnia based on network pharmacology, so as to further explore its mechanism of action. Methods: The chemical components of Chinese medicines in Zhizi Houpo decoction were collated by the traditional Chinese medicine systems pharmacology database and analysis platform and literature mining. The targets of chemical ingredients of Zhizi Houpo decoction were extracted from Pharm Mapper. Gene Cards, OMIM, Drugbank and TTD databases were used to collect related targets of depression, anxiety and insomnia. Venn tool was used to screen intersection targets. The protein-protein interaction(PPI) network of intersection targets was constructed using STRING database and Cytoscape 3.7.2 software to screen the core targets. Metascape was used for Go functional enrichment analysis and KEGG pathway enrichment analysis. A ingredient-target-pathway network was constructed by Cytoscape 3.7.2 software. Results: A total of 43 active ingredients of Zhizi Houpo decoction were screened out, and 290 corresponding targets were identified. The disease targets of depression, anxiety and insomnia were 1 323, 2 765 and 412, respectively. The number of disease-drug intersection targets predicted by Venn database was 137, 165 and 68, respectively. The core active ingredients were quercetin, kaempferol, luteolin, β-sitosterol, magnolithol, etc. KEGG enrichment showed that interleukin 17(IL-17), tumor necrosis factor(TNF), AGE-RAGE signaling pathway in diabetic complications were the main pathways in treating depression, anxiety and insomnia with Zhizi Houpo decoction. Conclusion: This study initially reveals that Zhizi Houpo decoction can treat depression, anxiety and insomnia through multiingredient, multi-target and multi-pathway, and provides a theoretical basis for in-depth clinical research of Zhizi Houpo decoction.
