摘 要:对不同萌发阶段的牛蒡种子(包括干种子,吸水24 h种子,培养4 d未发芽种子,幼苗,不发芽种子)进行内生菌的分离纯化、分子鉴定、功能测定、拮抗作用及牛蒡苷和牛蒡苷元含量变化分析。首先,通过平板划线法成功分离并纯化了21株可培养内生菌,主要优势菌为肠菌科,利用分子生物学技术明确了这些内生菌的分类地位及种类多样性,并对固氮、溶磷、产铁载体及产IAA的促生实验对内生菌功能进行探索,通过HPLC法分析内生菌影响不同萌发阶段牛蒡种子中牛蒡苷和牛蒡苷元的含量。同时,采用平板对峙法评估了内生菌对牛蒡炭疽病病原菌的拮抗作用,筛选出具有显著拮抗能力的菌株。最后,通过菌悬液处理种子的萌发实验,验证了这些内生菌对牛蒡生长的促进作用,提升了种子的发芽率和发芽势。本研究为牛蒡内生菌资源的开发利用及中药资源的可持续发展提供了科学依据和实践指导。This study was conducted to isolate,purify,and conduct molecular identification,functional assays,antagonistic effects,as well as analyze the changes in arctiin and arctigenin contents in different germination stages of Arctium lappa seeds,including dry seeds,seeds after 24-hour water absorption,seeds that did not germinate after 4-day culture,seedlings,and non-germinating seeds.Initially,21 culturable endophytic strains were successfully isolated and purified by the streak plate method,with Enterobacteriaceae being the predominant bacterial group.Using molecular biological techniques,the taxonomic status and species diversity of these endophytes were clarified.Furthermore,the functional exploration of these endophytes were conducted through experiments on nitrogen fixation,phosphate solubilization,siderophore production,and IAA synthesis.HPLC was used to analyze the influence of these endophytic on the arctiin and arctigenin content in A.lappa seeds across different germination stages.The content of arctiin and arctigenin in A.lappa seeds at different germination stages was analyzed using High-Performance Liquid Chromatography(HPLC)to assess the influence of endophytes.Meanwhile,the antagonistic effects of the endophytes against the anthracnose pathogen in A.lappa anthracnose were evaluated using the plate confrontation method,and strains with significant antagonistic capabilities were screened out.Finally,the germination experiment of treating seeds with bacterial suspension verified the promoting effect of these endophytes on the growth of A.lappa,and improved the germination rate and germination potential of seeds.This research provides scientific basis and practical guidance for the development and utilization of A.lappa endophytic resources,and the sustainable development of traditional Chinese medicine resources.
