摘 要:骨关节炎(OA)是一种慢性疾病,与软骨破坏、骨赘形成和滑膜炎症有关。长链非编码RNAs(lncRNAs)被广义地定义为长度超过200个核苷酸的非编码RNA分子,前沿研究表明,lncRNAs参与炎症性疾病、自身免疫病、癌症等多种疾病的发生机制,尤其在骨关节炎的软骨细胞分化、增殖、损伤和炎症反应等病理过程中具有重要作用。核因子kappa B(NF-κB)是一种转录因子,负责体内细胞的炎症、分化、增殖和存活等多种生物学功能,在OA中NF-κB通过桥接骨稳态和细胞炎症来加重OA。近年来,研究者发现lncRNAs可以通过调节NF-κB通路来影响OA的发生和进展。本综述总结不同的lncRNAs会通过哪些途径参与NF-κB通路从而影响OA,探讨lncRNAs促进NF-κB通路在OA中的意义,旨在提供更加明确的OA分子机制,OA研究的新思路和揭示未来治疗可能的潜在靶点。Osteoarthritis(OA)is a chronic disease associated with cartilage degradation,osteophyte formation,and synovial inflammation.Long non-coding RNAs(lncRNAs)are broadly defined as non-coding RNA molecules longer than 200 nucleotides.Emerging research suggests that lncRNAs are involved in the pathogenesis of inflammatory diseases,autoimmune diseases,cancer,and other conditions,particularly playing significant roles in the pathological processes of cartilage cell differentiation,proliferation,damage,and inflammatory responses in osteoarthritis.Nuclear factor kappa B(NF-κB)is a transcription factor responsible for a variety of biological functions such as inflammation,differentiation,proliferation and survival of cells in vivo,and NF-κB exacerbates OA by bridging bone homeostasis and cellular inflammation.In recent years,researchers have found that lncRNAs can affect the onset and progression of OA by regulating the NF-κB pathway.This review summarizes the pathways through which different lncRNAs will participate in the NF-κB pathway and thus affect OA,and also explores the significance of lncRNA promotion of the NF-κB pathway in OA,aiming to provide a clearer understanding of the molecular mechanisms of OA as well as new ideas for OA research,and reveal possible potential targets for future therapies.
